Increased myocyte apoptosis in diabetic hearts has been previously reported. The purpose of this study was to evaluate the effects of exercise training on cardiac survival pathways in streptozotocin (STZ)-induced diabetic rats.
Forty-eight male Wistar rats were randomly divided into control group (Control), STZ-induced (65 mg/kg, i.p.) diabetes (DM), and DM rats with moderate aerobic exercise training (DM-EX) on a treadmill 60 min/day, 5 days/week, for 10 weeks. Histopathological analysis, positive TUNEL assays and Western blotting were performed on the excised cardiac left ventricles from all three groups.
The components of cardiac survival pathway (insulin-like growth factor I (IGFI), IGFI-receptor (IGFI-R), phosphatidylinositol 3'-kinase (PI3K), and Akt) and the pro-survival Bcl-2 family proteins (Bcl-2, Bcl-xL, and p-BAD) were all significantly decreased in the DM group compared with the Control group whereas they were increased in the DM-EX group. In addition, the abnormal myocardial architecture, enlarged interstitial space and increased cardiac TUNEL-positive apoptotic cells were observed in the DM group, but they were reduced in the DM-EX group. The apoptotic key component, caspase-3, was significantly increased in the DM group relative to the Control group whereas it was decreased in the DM-EX group.
Exercise training enhances cardiac IGFI-R/PI3K/Akt and Bcl-2 family associated pro-survival pathways, which provides one of the new beneficial effects for exercise training in diabetes.

48 只雄性 Wistar 大鼠随机分为对照组(Control)、STZ 诱导的糖尿病组(DM)和 DM 大鼠进行中等强度有氧运动训练组(DM-EX),在跑步机上运动 60 分钟/天,每周 5 天,共 10 周。对三组大鼠切除的心脏左心室进行组织病理学分析、TUNEL 阳性检测和 Western blot 分析。
与对照组相比,DM 组心脏存活途径的组成部分(胰岛素样生长因子 I(IGFI)、IGFI 受体(IGFI-R)、磷脂酰肌醇 3'-激酶(PI3K)和 Akt)和促生存 Bcl-2 家族蛋白(Bcl-2、Bcl-xL 和 p-BAD)均显著降低,而 DM-EX 组则增加。此外,在 DM 组中观察到异常的心肌结构、扩大的间质空间和增加的心脏 TUNEL 阳性凋亡细胞,但在 DM-EX 组中减少。凋亡关键成分 caspase-3 在 DM 组中相对于对照组显著增加,而在 DM-EX 组中降低。
运动训练增强了心脏 IGFI-R/PI3K/Akt 和 Bcl-2 家族相关的促生存途径,为糖尿病患者运动训练提供了新的有益效果之一。
