To investigate the reliability of magnetic-activated cell sorting (MACS) combined with multiplex ligation-dependent probe amplification (MLPA) in the detection of the molecular cytogenetic abnormalities in multiple myeloma.
The bone marrow cells were collected from 29 patients with multiple myeloma. The immuno magnetically sorted and unsorted cells were detected for TP53 and RB1 expressions using MLPA probes and the results were compared with fluorescent in situ hybridization (FISH).
The detection success rate was 100% for MLPA, which yielded results with an concordance rate of 99.1% with the FISH results. The positivity rates of MLPA and FISH were both increased after immunomagnetic sorting of the bone marrow cells.
MLPA can well suit the clinical needs for detecting molecular cytogenetic abnormalities in multiple myeloma, and the samples should be immuno magnetically sorted before the assay.
探讨磁珠分选法(MACS)联合多重连接依赖探针扩增技术(MLPA)检测多发性骨髓瘤分子细胞遗传学异常的可靠性。
收集29例多发性骨髓瘤患者的骨髓细胞。使用MLPA探针检测免疫磁珠分选前后细胞中TP53和RB1的表达,并将结果与荧光原位杂交(FISH)结果进行比较。
MLPA检测成功率为100%,与FISH结果的一致性率为99.1%。骨髓细胞免疫磁珠分选后,MLPA和FISH的阳性率均升高。
MLPA非常适合多发性骨髓瘤分子细胞遗传学异常的临床检测需求,检测前应对样本进行免疫磁珠分选。
