To explore the expression pattern of microRNA (miRNA) in T cells of peripheral blood mononuclear cell (PBMC) from patients with primary biliary cirrhosis (PBC).
The expression profile of miRNA in T cells of PBMC was determined by microarray assay and validated by real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR).
In comparison with the healthy controls, 23 miRNA were down-regulated and 2 miRNA had a higher expression (all P < 0.05). As revealed by qRT-PCR, the expressions of miR-346, miR-17-5p, miR-20a and miR-let-7b decreased obviously while miR-451 and miR-129 became up-regulated. The results were in agreement with those of microarray.
The PBC patients and healthy controls have significantly different expression profiles of microRNA in T cells of PBMC. The differential expression of microRNA may be involved in the pathogenesis of PBC.
探讨原发性胆汁性肝硬化(PBC)患者外周血单个核细胞(PBMC)中T细胞微小RNA(miRNA)的表达模式。
采用基因芯片技术检测PBMC中T细胞miRNA的表达谱,并通过实时定量逆转录聚合酶链反应(qRT-PCR)进行验证。
与健康对照相比,23种miRNA表达下调,2种miRNA表达上调(均P < 0.05)。qRT-PCR结果显示,miR-346、miR-17-5p、miR-20a和miR-let-7b表达明显降低,而miR-451和miR-129表达上调。结果与基因芯片结果一致。
PBC患者与健康对照PBMC中T细胞的miRNA表达谱存在显著差异。miRNA的差异表达可能参与PBC的发病机制。
