Abstract

OBJECTIVE

In order to explore the diversity, antimicrobial activity and enzyme-producing activity of marine actinobacteria isolated from mangrove sediments in Indian Ocean.

METHODS

Eight sediments collected from mangrove sediments in Indian Ocean were treated by the plate dilution method and spread on 24 isolation media only containing sole carbon source for energy. Marine actinobacteria were isolated and identified by 16S rRNA gene sequence analysis. The antimicrobial activity and enzyme-producing activity of isolated strains were further detected by spot planting method.

RESULTS

In total 139 representative strains were selected from 521 isolates, and they were further sequenced and performed phylogenetic analysis based on their 16S rRNA gene sequences. There were 35 strains identified as potential novel species. Antimicrobial activity was detected in Bacillus subtilis, Candida albicans, Escherichia coli, Staphylococcus aureus, Aspergillus niger. Enzyme-producing activity for protease cellulase, amylase and esterase were 36.5%, 26.5%, 22.4% and 15.9%, respectively.

CONCLUSION

Diverse marine actinobacteria were discovered in mangrove sediment in Indian Ocean, which have antimicrobial and enzyme activity.

目的

为了探索从印度洋红树林沉积物中分离出的海洋放线菌的多样性、抗菌活性和产酶活性。

方法

采用平板稀释法处理从印度洋红树林沉积物中采集的8份沉积物,并将其涂布在仅含单一碳源作为能源的24种分离培养基上。通过16S rRNA基因序列分析对海洋放线菌进行分离和鉴定。采用点种法进一步检测分离菌株的抗菌活性和产酶活性。

结果

从521株分离物中总共筛选出139株代表性菌株,并基于其16S rRNA基因序列进行进一步测序和系统发育分析。有35株被鉴定为潜在的新物种。在枯草芽孢杆菌、白色念珠菌、大肠杆菌、金黄色葡萄球菌、黑曲霉中检测到抗菌活性。蛋白酶、纤维素酶、淀粉酶和酯酶的产酶活性分别为36.5%、26.5%、22.4%和15.9%。

结论

在印度洋红树林沉积物中发现了多种具有抗菌和酶活性的海洋放线菌。
