This study aimed to investigate the expression of B7-H4 in human thyroid cancer and determine any association with patient clinicopathological parameters and survival.
B7-H4 expression in 64 clinical thyroid cancer specimens was assessed with immunohistochemistry. Moreover, B7-H4 mRNA expression in 10 fresh resected specimens were evaluated by the reverse transcription-polymerase chain reaction (RT-PCR). Immunohistochemical staining of CD3 was performed to assess the number of tumor infiltrating T lymphocytes (TILs) in thyroid cancers.
Positive B7-H4 immunohistochemical staining was observed in 61 out of 64 (95.3%) specimens of thyroid cancer tissues. Significantly more B7-H4 mRNA copies were found in thyroid cancer tissue than that adjacent normal tissue. Moreover, B7-H4 expression in human thyroid cancer tissues was significantly correlated with patient TNM stages and extrathyroidal extension (P<0.05), being inversely correlated with the number of TILs (P<0.05). The overall survival rate of the patients with higher B7-H4 expression was significantly worse than that of the patients with lower B7-H4 expression.
This present study suggests that high B7-H4 expression is associated with cancer progression, reduced tumor immunosurveillance and worse patient outcomes in human thyroid cancer.
本研究旨在调查B7-H4在人甲状腺癌中的表达情况,并确定其与患者临床病理参数及生存情况之间的关联。
采用免疫组织化学方法评估64例临床甲状腺癌标本中B7-H4的表达。此外,通过逆转录-聚合酶链反应(RT-PCR)评估10例新鲜切除标本中B7-H4 mRNA的表达。进行CD3免疫组织化学染色以评估甲状腺癌中肿瘤浸润性T淋巴细胞(TILs)的数量。
在64例甲状腺癌组织标本中的61例(95.3%)观察到B7-H4免疫组织化学染色呈阳性。甲状腺癌组织中发现的B7-H4 mRNA拷贝数明显多于相邻正常组织。此外,人甲状腺癌组织中B7-H4的表达与患者TNM分期和甲状腺外侵犯显著相关(P<0.05),与TILs数量呈负相关(P<0.05)。B7-H4表达较高的患者总生存率明显低于B7-H4表达较低的患者。
本研究表明,B7-H4高表达与人甲状腺癌的肿瘤进展、肿瘤免疫监视降低及患者预后较差有关。
