Abstract

BACKGROUND AND OBJECTIVE

The survival rate of lung cancer is low, thus new methods for treating this form of cancer must be explored. This study applies immune therapy with metronomic chemotherapy to observe the effect of combined therapy on suppressing tumor.

METHODS

Mice were inoculated with Lewis lung carcinoma cells. Different treatments, namely, saline, metronomic chemotherapy, dendritic cell (DC) vaccine, and metronomic chemotherapy with DC vaccine were administered to corresponding mice groups. Basic fibroblast growth factor and vascular endothelial growth factor (VEGF) were detected via microdialysis and Luminex.

RESULTS

The median survival time of mice in the metronomic chemotherapy with DC vaccine group was (27.6±3.2) days, whereas that in the saline group was (13.5±2.7) days (P=0.008), that in the DC) vaccine group was (13.1±2.3) days (P=0.01), and that in the metronomic chemotherapy group was (11.8±3.0) days (P=0.01). The mice in the metronomic chemotherapy with DC vaccine group exhibited longer survival time than the mice in the other groups. This result could be related to the downregulation of VEGF secretion in the tumor-bearing mice groups within 48 h to 72 h.

CONCLUSIONS

VEGF secretion could be downregulated in tumor-bearing mice by administering metronomic chemotherapy with DC vaccine.

背景与目的

肺癌生存率较低,因此必须探索治疗这种癌症的新方法。本研究应用节拍化疗免疫疗法观察联合治疗对抑制肿瘤的效果。

方法

给小鼠接种Lewis肺癌细胞。对相应小鼠组给予不同治疗,即生理盐水、节拍化疗、树突状细胞(DC)疫苗以及DC疫苗联合节拍化疗。通过微透析和Luminex检测碱性成纤维细胞生长因子和血管内皮生长因子(VEGF)。

结果

DC疫苗联合节拍化疗组小鼠的中位生存时间为(27.6±3.2)天,而生理盐水组为(13.5±2.7)天(P = 0.008),DC疫苗组为(13.1±2.3)天(P = 0.01),节拍化疗组为(11.8±3.0)天(P = 0.01)。DC疫苗联合节拍化疗组小鼠的生存时间比其他组小鼠长。这一结果可能与荷瘤小鼠组在48小时至72小时内VEGF分泌下调有关。

结论

通过给予DC疫苗联合节拍化疗可使荷瘤小鼠的VEGF分泌下调。
