To investigate the effect of perinatal high fat diet on spatial learning and memory in adult Kunming (KM) mice offspring.
The adult female KM mice were randomly divided into control and high fat group, fed with lab chow and high fat diet (containing 20% of lard) during gestation and lactation periods. After weaning at 4 weeks old, 10 offspring mice randomly selected from control group, as control continuing (CC) group, were fed with lab chow. 10 mice, as high fat continuing (FF) group, from high fat group were fed with high fat diet. 10 mice, as transforming high fat (FC) group, were fed with lab chow. Then, Morris water maze task was applied to evaluate spatial learning and memory in the pups. The body weight and the mass of visceral fat and brain were weighted, and the level of serum leptin was measured through ELISA.
Latency of FF and FC groups were shorter than CC group (P < 0.05), and the crossing times of FC group was more than CC group. The weights, the mass of visceral fat and brain of FF and FC groups were heavier than those of control group (P < 0.05). The level of serum leptin of FF group was higher than CC group (P <0. 05).
Maternal high fat diet during pregnancy and lactation may contribute to better spatial learning and memory ability in adult mice offspring.
探讨围产期高脂饮食对成年昆明(KM)小鼠子代空间学习和记忆的影响。
将成年雌性KM小鼠随机分为对照组和高脂组,在妊娠和哺乳期分别给予普通饲料和高脂饲料(含20%猪油)。4周龄断奶后,从对照组随机选取10只子代小鼠作为继续对照组(CC组),给予普通饲料。从高脂组选取10只小鼠作为高脂继续组(FF组),给予高脂饲料。选取10只小鼠作为高脂转换组(FC组),给予普通饲料。然后,采用Morris水迷宫任务评估幼崽的空间学习和记忆能力。称量体重、内脏脂肪和脑质量,并通过酶联免疫吸附测定法测量血清瘦素水平。
FF组和FC组的潜伏期短于CC组(P < 0.05),且FC组的穿越次数多于CC组。FF组和FC组的体重、内脏脂肪和脑质量均高于对照组(P < 0.05)。FF组的血清瘦素水平高于CC组(P < 0.05)。
孕期和哺乳期母体高脂饮食可能有助于成年小鼠子代具有更好的空间学习和记忆能力。
