Abstract

AIMS

Early diagnosis is important for cervical cancer treatment. This study aimed to characteriz the microRNA profile and target gene protein levels of cervical cancers in Uygur women for application in early diagnosis.

METHODS

The profiles of miRNA in cervical cancer and chronic cervicitis were analyzed with miRNAmicroarray V4.0. The expression of miR-101 was detected by real-time PCR and locked nucleotide acid in situ hybridization (LNA-ISH). Cox-2 protein levels were assessed by immunohistochemistry.

RESULTS

The microarray identified a set of 12 miRNAs significantly decreased in cervical cancer in comparison to the control group. Quantitative RT-PCR analysis showed miR-101 to be significantly downregulated in cancer tissues (p<0.05) while LNA-ISH showed miR-101 positive rates of 80% (20/25) and 8% (5/25) (p<0.05) in the control and cervical cancer groups. Cox-2 positive rates of cervical cancer and control groups were 84% (21/25) and 8% (2/25) (p<0.05).

CONCLUSIONS

Use of down-regulation of miR-101 and up-regulation of Cox-2 as markers may play a role in early diagnosis of cervical cancer in Uygur women.

目的

早期诊断对宫颈癌治疗至关重要。本研究旨在描绘维吾尔族女性宫颈癌的微小RNA图谱及靶基因蛋白水平,以应用于早期诊断。

方法

采用miRNA微阵列V4.0分析宫颈癌和慢性宫颈炎中的miRNA图谱。通过实时聚合酶链反应和锁核酸原位杂交(LNA-ISH)检测miR-101的表达。采用免疫组织化学评估Cox-2蛋白水平。

结果

与对照组相比,微阵列鉴定出一组在宫颈癌中显著降低的12种miRNA。定量逆转录聚合酶链反应分析显示,癌组织中miR-101显著下调(p<0.05),而LNA-ISH显示对照组和宫颈癌组中miR-101阳性率分别为80%(20/25)和8%(5/25)(p<0.05)。宫颈癌组和对照组的Cox-2阳性率分别为84%(21/25)和8%(2/25)(p<0.05)。

结论
