To explore the effect of adipose-derived stem cells (ADSCs) transplantation on the expression of neurite outgrowth inhibitor-A (Nogo-A) after cerebral ischemia in middle cerebral artery occlusion (MCAO) rats.
ADSCs were harvested and purified, and then MCAO models were established in rats by the suture method. Rats were divided into sham operation group, MCAO model group and ADSCs-injected group. One day after MCAO, ADSCs were injected into the lateral ventricle of the ADSC group rats and the same does of PBS was given to model group rats. At day 4, 7 and 14 after MCAO, the rats were decapitated to detect the Nogo-A expression in the perilesional cortex by reverse transcription PCR, immunohistochemistry and Western blotting.
Compared with sham group, the expression of Nogo-A was significantly up-regulated at each time point in both model group and ADSC group. Compared with model group, the expression of Nogo-A was significantly down-regulated at each time point in ADSC group.
ADSC transplantation could promote the recovery of neurological functions, partly by inhibiting the expression of Nogo-A in MCAO rat's brain.
探讨脂肪来源干细胞(ADSCs)移植对大脑中动脉闭塞(MCAO)大鼠脑缺血后神经突生长抑制因子-A(Nogo-A)表达的影响。
获取并纯化ADSCs,然后通过缝合方法在大鼠中建立MCAO模型。将大鼠分为假手术组、MCAO模型组和ADSCs注射组。MCAO术后1天,将ADSCs注入ADSC组大鼠的侧脑室,模型组大鼠给予相同剂量的PBS。在MCAO术后第4、7和14天,将大鼠断头,通过逆转录PCR、免疫组织化学和蛋白质印迹法检测损伤周围皮质中Nogo-A的表达。
与假手术组相比,模型组和ADSC组在各个时间点Nogo-A的表达均显著上调。与模型组相比,ADSC组在各个时间点Nogo-A的表达均显著下调。
ADSC移植可促进神经功能恢复,部分原因是抑制MCAO大鼠脑中Nogo-A的表达。
