To investigate the expression of vascular endothelial growth factor (VEGF) and hypoxia-inducible factor 1α (HIF-1α) in infarct area of rats with acute myocardial infarction (AMI).
A total of 36 healthy male Wistar rats weighing 180 g-220 g were included in our study and were randomly divided into two groups of 6 rats each: sham operation group and experiment group. In sham operation group, surgery was performed by opening chest without ligation of arteria coronaria while, in the experiment group, surgery was performed to produce AMI model. Animals were sacrificed immediately after the operation on day 7 and day 14, respectively. The serum troponin, myocardial infarct area, microvessel density in infarct area, VEGF and HIF-1α expression were analyzed.
Differences in the serum troponin level, myocardial infarct area, microvessel density in infarct area, VEGF and HIF-1 expression level at different time points in sham and experiment groups had statistical significance (p < 0.05). On day 7, the serum troponin, myocardial infarct area, microvessel density in infarct area, VEGF and HIF-1 expression level were the highest and the level was second highest on day 14 while the levels were lowest immediately after the operation. The expression levels of VEGF and HIF-1α were positively related with the increasing density of microvessel in infarct area (p < 0.05).
The expression of VEGF and HIF-1α might be involved with myocardial remodeling and angiogenesis.
探讨血管内皮生长因子(VEGF)和缺氧诱导因子1α(HIF-1α)在急性心肌梗死(AMI)大鼠梗死区域的表达情况。
选取36只体重180 g - 220 g的健康雄性Wistar大鼠纳入本研究,随机分为两组,每组6只:假手术组和实验组。假手术组仅打开胸腔但不结扎冠状动脉进行手术,而实验组进行手术以建立AMI模型。分别在术后第7天和第14天立即处死动物,分析血清肌钙蛋白、心肌梗死面积、梗死区域微血管密度、VEGF和HIF-1α的表达情况。
假手术组和实验组在不同时间点的血清肌钙蛋白水平、心肌梗死面积、梗死区域微血管密度、VEGF和HIF-1表达水平差异具有统计学意义(p < 0.05)。术后第7天,血清肌钙蛋白、心肌梗死面积、梗死区域微血管密度、VEGF和HIF-1表达水平最高,术后第14天次之,术后即刻最低。VEGF和HIF-1α的表达水平与梗死区域微血管密度增加呈正相关(p < 0.05)。
VEGF和HIF-1α的表达可能参与心肌重塑和血管生成。
