To investigate the alteration in rat temporomandibular joint (TMJ) synovial membrane induced by increased occlusal vertical dimension (iOVD) and to determine whether the p38 mitogen activated protein kinase (MAPK) signaling cascade is involved.
Thirty-six rats were randomly divided into 3 groups: control + normal saline (NS; controls), iOVD + NS, and iOVD + SB203580 (a potent p38 MAPK inhibitor). Morphologic changes of synovial tissues were observed and scored. Activation levels of p38 MAPK and activating transcription factor-2 (ATF2) were detected by immunohistochemistry. Expression levels of interleukin-1β (IL-1β) and matrix metalloproteinase-3 (MMP-3) were measured by quantitative real-time polymerase chain reaction and immunohistochemistry.
Obvious synovitis was found in the iOVD group. P38 and ATF2 were activated, and mRNA and protein expression levels of IL-1β and MMP-3 were upregulated after iOVD. However, decreased synovial tissue inflammation and lower mRNA and protein levels of IL-1β and MMP-3 were observed in the iOVD + SB203580 group.
iOVD can induce temporomandibular joint synovitis, and the p38 MAPK signaling cascade might participate in and aggravate the process of articular inflammation.

36只大鼠随机分为3组:对照组 + 生理盐水(NS;对照组)、iOVD + NS组和iOVD + SB203580组(一种有效的p38 MAPK抑制剂)。观察滑膜组织的形态学变化并进行评分。通过免疫组织化学检测p38 MAPK和激活转录因子-2(ATF2)的激活水平。通过定量实时聚合酶链反应和免疫组织化学测量白细胞介素-1β(IL-1β)和基质金属蛋白酶-3(MMP-3)的表达水平。
iOVD组发现明显的滑膜炎。iOVD后p38和ATF2被激活,IL-1β和MMP-3的mRNA和蛋白表达水平上调。然而,在iOVD + SB203580组中观察到滑膜组织炎症减轻,IL-1β和MMP-3的mRNA和蛋白水平降低。
iOVD可诱导颞下颌关节滑膜炎,p38 MAPK信号级联可能参与并加重关节炎症过程。
