This study was designed to investigate the expression of regulatory T cells in primary Sjögren's Syndrome (pSS) and to evaluate the clinical role of CD4 Helios FoxP3 cells in pSS patients.
CD4 FoxP3T cells in the peripheral blood of 39 pSS patients and 30 healthy controls were measured by flow cytometry and CD25 and Helios expression were also analyzed. The repression ability of CD4 CD25 cells was tested in vitro. Clinical information of pSS patients was retrospectively collected and their correlations with circulating Treg cells were analyzed. Cytokine levels in plasma were measured by ELISA and correlations with Helios FoxP3cells were also detected.
Circulating FoxP3and Helios FoxP3cells were elevated in pSS patients compared with controls. The suppression function of CD4 CD25 cells is not different between two groups. There are inverse correlations between Helios FoxP3percentage and ESR, IgG, IgM and ESSDAI. Anti-SSB patients possess higher level of Helios FoxP3cells than anti-SSBpatients. IL-6, IFNγ and IFNα levels were increased in pSS plasma and there were positive correlations between the levels of IFNγ/IFNα and percentage of Helios FoxP3cells.
Circulating Helios FoxP3 cells were elevated in pSS patients and may contribute to suppressing autoimmunity in pSS patients.
本研究旨在调查原发性干燥综合征(pSS)中调节性T细胞的表达,并评估CD4⁺Helios⁺FoxP3细胞在pSS患者中的临床作用。
采用流式细胞术检测39例pSS患者和30例健康对照外周血中CD4⁺FoxP3⁺T细胞,并分析CD25和Helios的表达。体外检测CD4⁺CD25⁺细胞的抑制能力。回顾性收集pSS患者的临床资料,并分析其与循环Treg细胞的相关性。采用ELISA法检测血浆细胞因子水平,并检测其与Helios⁺FoxP3⁺细胞的相关性。
与对照组相比,pSS患者循环中FoxP3⁺和Helios⁺FoxP3⁺细胞升高。两组CD4⁺CD25⁺细胞的抑制功能无差异。Helios⁺FoxP3⁺百分比与ESR、IgG、IgM和ESSDAI呈负相关。抗SSB患者的Helios⁺FoxP3⁺细胞水平高于抗SSA患者。pSS血浆中IL-6、IFNγ和IFNα水平升高,IFNγ/IFNα水平与Helios⁺FoxP3⁺细胞百分比呈正相关。
pSS患者循环中Helios⁺FoxP3⁺细胞升高,可能有助于抑制pSS患者的自身免疫。
