To investigate patterns of action potential firing in cortical heurons of neonatal mice and their electrophysiological properties.
The passive and active membrane properties of cortical neurons from 3-d neonatal mice were observed by whole-cell patch clamp with different voltage and current mode.
Three patterns of action potential firing were identified in response to depolarized current injection. The effects of action potential firing patterns on voltage-dependent inward and outward current were found. Neurons with three different firing patterns had different thresholds of depolarized current. In the morphology analysis of action potential, the three type neurons were different in rise time, duration, amplitude and threshold of the first action potential evoked by 80 pA current injection. The passive properties were similar in three patterns of action potential firing.
These results indicate that newborn cortical neurons exhibit different patterns of action potential firing with different action potential parameters such as shape and threshold.
研究新生小鼠皮质神经元动作电位发放模式及其电生理特性。
采用全细胞膜片钳技术,在不同电压和电流模式下观察3日龄新生小鼠皮质神经元的被动和主动膜特性。
在去极化电流注入时可识别出三种动作电位发放模式。发现了动作电位发放模式对电压依赖性内向和外向电流的影响。具有三种不同发放模式的神经元去极化电流阈值不同。在动作电位的形态学分析中,三种类型的神经元在由80 pA电流注入诱发的第一个动作电位的上升时间、持续时间、幅度和阈值方面存在差异。三种动作电位发放模式的被动特性相似。
这些结果表明,新生皮质神经元表现出不同的动作电位发放模式,具有不同的动作电位参数,如形状和阈值。
