Cadmium is one of the potent environmental endocrine disruptors that has adverse effects on male fertility. This study was conducted to investigate the protective role of green tea extract (GTE) on cadmium chloride (CdCl) induced reproductive toxicities in male Wistar rats. Thirty-six male Wistar rats were divided to 4 groups, each one contained 9 rats. Control group, they were gavaged distilled water, meanwhile the treated groups gavaged CdCl (3mg/kg), GTE (70mg/kg) and CdCl+GTE (3mg/kg and 70mg/kg) for 63days. In GTE+ CdCl treated rats, the final body weight, relative testicular weight, epididymal weight, seminal and prostate glands weights were significantly (P<0.05) increased compared to CdCl intoxicated rats. Sperm cell concentration was significantly (P<0.05) increased in CdCl+GTE group, while sperm morphological abnormalities were significantly (P<0.05) decreased than CdCl group. The levels of superoxide dismutase (SOD) and reduced glutathione (GSH) were elevated significantly (P<0.05) in CdCl+GTE group, meanwhile catalase activity was significantly (P<0.05) declined than CdCl group. Hypercholesterolemia was observed in CdCl group than control one while GTE+CdCl group revealed significant (P<0.05) reduction in cholesterol level than CdCl treated group. Testicular androgen receptors and caspase-3 protein expression showed marked reduction and increase (P<0.05) in CdCl group than control, respectively. Treatment with GTE with CdCl significantly (P<0.05) increased androgen receptors, while decreased caspase-3 protein than CdCl group. In conclusion, GTE exhibited protective effect on testes by inhibiting CdCl induced oxidative damage and cellular apoptosis.
镉是一种强效的环境内分泌干扰物,对男性生育能力有不良影响。本研究旨在探讨绿茶提取物(GTE)对氯化镉(CdCl)诱导的雄性Wistar大鼠生殖毒性的保护作用。将36只雄性Wistar大鼠分为4组,每组9只。对照组给予蒸馏水灌胃,同时治疗组分别给予CdCl(3mg/kg)、GTE(70mg/kg)以及CdCl+GTE(3mg/kg和70mg/kg)灌胃63天。在GTE+CdCl处理的大鼠中,与CdCl中毒大鼠相比,最终体重、相对睾丸重量、附睾重量、精囊和前列腺重量均显著(P<0.05)增加。CdCl+GTE组精子细胞浓度显著(P<0.05)增加,而精子形态异常率比CdCl组显著(P<0.05)降低。CdCl+GTE组超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)水平显著(P<0.05)升高,同时过氧化氢酶活性比CdCl组显著(P<0.05)下降。与对照组相比,CdCl组出现高胆固醇血症,而GTE+CdCl组胆固醇水平比CdCl处理组显著(P<0.05)降低。睾丸雄激素受体和半胱天冬酶-3蛋白表达在CdCl组与对照组相比分别显著降低和增加(P<0.05)。GTE与CdCl联合处理显著(P<0.05)增加雄激素受体,同时比CdCl组降低半胱天冬酶-3蛋白表达。总之,GTE通过抑制CdCl诱导的氧化损伤和细胞凋亡对睾丸表现出保护作用。
