Abstract

The purpose of this study was to delineate the various factors that affect the growth characteristics of human cancer xenografts in nude mice and to reveal the relationship between the growth characteristics and radiosensitivity. We retrospectively analyzed 390 xenografts comprising nine different human cancer lines grown in nude mice used in our institute between 2009 and 2015. Tumor growth rate (TGR) was calculated using exponential growth equations. The relationship between the TGR of xenografts and the proliferation of the cells was examined. Additionally, we examined the correlations between the surviving fractions of cells after 2 Gy irradiation and the response of the xenograft to radiation. The TGR of xenografts was positively related to the proliferation of the cells ( =0.9714, <0.0001), whereas it was independent of the histological type of the xenografts. Radiation-induced suppression of the growth rate (T/C%) of xenografts was positively related to the radiosensitivity of the cells (SF; =0.8684, =0.0284) and TGR ( =0.7623, =0.0780). The proliferation of human cancer cells and the growth rate of xenografts were positively related. The radiosensitivity of cancer cells, as judged from the SF values , and the radiation-induced suppression of xenograft growth were positively related. In conclusion, the growth rate of human xenografts was independent of histological type and origin of the cancer cells, and was positively related to the proliferation of the cancer cells .

本研究的目的是阐明影响人癌异种移植瘤在裸鼠体内生长特征的各种因素,并揭示生长特征与放射敏感性之间的关系。我们回顾性分析了2009年至2015年间在我院用于裸鼠体内生长的包含9种不同人癌细胞系的390个异种移植瘤。使用指数生长方程计算肿瘤生长率(TGR)。研究了异种移植瘤的TGR与细胞增殖之间的关系。此外,我们还研究了2 Gy照射后细胞存活分数与异种移植瘤对辐射反应之间的相关性。异种移植瘤的TGR与细胞增殖呈正相关(r = 0.9714,P < 0.0001),而与异种移植瘤的组织学类型无关。辐射诱导的异种移植瘤生长率抑制(T/C%)与细胞放射敏感性(SF;r = 0.8684,P = 0.0284)和TGR(r = 0.7623,P = 0.0780)呈正相关。人癌细胞增殖与异种移植瘤生长率呈正相关。根据SF值判断的癌细胞放射敏感性与辐射诱导的异种移植瘤生长抑制呈正相关。总之,人异种移植瘤的生长率与癌细胞的组织学类型和来源无关,而与癌细胞增殖呈正相关。
