Abstract

OBJECTIVE

To assess the effect of bovine lactoferrin (bLf) on the invasion of Salmonella enterica ser. Typhimurium to HEp-2 cells.

MATERIALS AND METHODS

HEp-2 monolayers were infected with 106 colony forming unit (CFU) of bacteria in the absence and presence of 1 and 10 mg/mL of bLf (iron-saturated) and incubated 1.5 hours at 37°C. Two treatments were evaluated: pre- infection (HEp-2 cells were incubated with bLf one hour prior to infection with Salmonella) and post-infection (bLf was added 15 minutes after the infection). Invasiveness of Salmonella was determined throgh quantification of CFU recovered from inside the HEp-2 cells (after treatment with 100 μg/mL and 10 μg/mL of gentamicin and Triton X -100).

RESULTS

In the pre-infection treatment, we observed a decrease of 23% of Salmonella invasion when HEp-2 cells were pre incubated with 1 mg/mL of bLf (2.8x105 vs 2.1x105, p=0.04) and 50% when them were pre-incubated with 10 mg/mL of bLf (2.8x105 vs 1.4x105, p=0.04). In post-infection treatment, no changes were observed in the invasiveness of Salmonella.

CONCLUSION

The results indicated that bLf reduces the invasiveness of Salmonella enterica ser. Typhimurium to HEp-2 cells in the pre-infection treatment.

目的

评估牛乳铁蛋白(bLf)对鼠伤寒沙门氏菌侵袭人喉表皮样癌细胞(HEp-2细胞)的影响。

材料与方法

在不存在和存在1 mg/mL及10 mg/mL(铁饱和)的bLf的情况下,用10⁶个菌落形成单位(CFU)的细菌感染HEp-2细胞单层,并于37°C孵育1.5小时。评估两种处理方式:感染前(在感染沙门氏菌前1小时将HEp-2细胞与bLf孵育)和感染后(感染后15分钟添加bLf)。通过对从HEp-2细胞内回收的CFU进行定量(在用100 μg/mL和10 μg/mL庆大霉素及曲拉通X-100处理后)来确定沙门氏菌的侵袭性。

结果

在感染前处理中,当用1 mg/mL的bLf预孵育HEp-2细胞时,我们观察到沙门氏菌侵袭减少了23%(2.8×10⁵对2.1×10⁵,p = 0.04),当用10 mg/mL的bLf预孵育时减少了50%(2.8×10⁵对1.4×10⁵,p = 0.04)。在感染后处理中,未观察到沙门氏菌侵袭性的变化。

结论

结果表明,在感染前处理中,bLf可降低鼠伤寒沙门氏菌对HEp-2细胞的侵袭性。
