Abstract

Previous research found that the polysaccharides isolated from Hohenbuehelia serotina possessed various biological activities, such as antioxidant, immunomodulation and radioprotective effects. However, the structural information of H. serotina polysaccharides has not yet been reported. Therefore, based on the investigation of the antioxidative tracking, a novel polysaccharide named as NTHSP-A1 was isolated from H. serotina by ultrasonic-assistance extraction and anion-exchange and gel permeation chromatography approaches. Structural characterization revealed that NTHSP-A1 had an average molecular weight of 8.09×10Da and composed of arabinose, mannose, glucose and galactose in a molar ratio of 4:16:28:11. The polysaccharide was semi-crystalline substance with multi-branching structure. The backbone of NTHSP-A1 was shown to contain →3,6)-α-d-Glcp-(1→, with branches substituted at C-3 of →2)-α-l-Arap-(1→, C-3 of α-d-Manp-(1→, and C-6 of →6)-β-d-Galp-(1→, respectively. This study provides a theoretical basis for the further research on the relationship between biological activity and structure of NTHSP-A1.

先前的研究发现,从晚生脐菇中分离出的多糖具有多种生物活性,如抗氧化、免疫调节和辐射防护作用。然而,晚生脐菇多糖的结构信息尚未见报道。因此,基于抗氧化追踪研究,通过超声辅助提取、阴离子交换和凝胶渗透色谱法从晚生脐菇中分离出一种名为NTHSP-A1的新型多糖。结构表征显示,NTHSP-A1的平均分子量为8.09×10Da,由阿拉伯糖、甘露糖、葡萄糖和半乳糖以4:16:28:11的摩尔比组成。该多糖为具有多分支结构的半结晶物质。NTHSP-A1的主链显示含有→3,6)-α-d-葡萄糖-(1→,分支分别取代于→2)-α-l-阿拉伯糖-(1→的C-3、α-d-甘露糖-(1→的C-3和→6)-β-d-半乳糖-(1→的C-6处。本研究为进一步研究NTHSP-A1的生物活性与结构之间的关系提供了理论依据。
