Abstract

The purpose of this study was to investigate whether the use of HLA as an aqueous binder of hydroxyapatite/beta-tricalcium phosphate (HA-TCP) particles can reduce the amount of bone graft needed and increase ease of handling in clinical situations. In this study, HA/TCP was loaded in commercially available crosslinking HLA to form a novel HLA/HA-TCP composite. Six New Zealand White rabbits (3.0-3.6 kg) were used as test subjects. Four 6 mm defects were prepared in the parietal bone. The defects were filled with the HLA/HA-TCP composite as well as HA-TCP particle alone. New bone formation was analyzed by micro-CT and histomorphometry. Our results indicated that even when the HA-TCP particle numbers were reduced, the regenerative effect on bone remained when the HLA existed. The bone volume density (BV/TV ratio) of HLA/HA-TCP samples was 1.7 times larger than that of the control sample at week 2. The new bone increasing ratio (NBIR) of HLA/HA-TCP samples was 1.78 times higher than the control group at week 2. In conclusion, HA-TCP powder with HLA contributed to bone healing in rabbit calvarial bone defects. The addition of HLA to bone grafts not only promoted osteoconduction but also improved handling characteristics in clinical situations.

本研究的目的是调查使用人白细胞抗原(HLA)作为羟基磷灰石/β-磷酸三钙(HA-TCP)颗粒的水性粘合剂是否可以减少所需骨移植材料的用量,并提高临床应用中的操作便利性。在本研究中,将HA/TCP负载于市售的交联HLA中,以形成新型的HLA/HA-TCP复合材料。选用6只体重3.0 - 3.6千克的新西兰白兔作为试验对象。在顶骨制备4个6毫米的缺损。缺损分别用HLA/HA-TCP复合材料以及单独的HA-TCP颗粒填充。通过显微CT和组织形态计量学分析新骨形成情况。我们的结果表明,即使减少HA-TCP颗粒的数量,当存在HLA时,对骨的再生效果依然存在。在第2周时,HLA/HA-TCP样本的骨体积密度(BV/TV比值)比对照组样本大1.7倍。在第2周时,HLA/HA-TCP样本的新骨增加率(NBIR)比对照组高1.78倍。总之,含有HLA的HA-TCP粉末有助于兔颅骨缺损的骨愈合。在骨移植材料中添加HLA不仅促进了骨传导,还改善了临床应用中的操作特性。
