Abstract

Bone marrow derived-mesenchymal stem cells (BM-MSCs) have brought great attention in regenerative medicine field, various experimental & clinical trials were held to investigate their therapeutic effects in different disorders. We designed a histological & immunohistochemical study to evaluate effectiveness of MSCs therapy in withhold of end-stage renal disease (ESRD) secondary to hypertension which has become a growing & striking public health problem. 30 adult male albino rats were utilized, 20 of them were exposed to experimental induction of hypertension, then divided equally to MSCs treated group (injected with 1×10 fluorescent labeled cell i.v./rat), while the second one was left without treatment. Renal specimens were subjected to histopathological, ultrastructural and immunohistochemical examination for Nrf2 in addition to biochemical estimation of serum urea & creatinine. Our results documented that BM-derived MSCs exerts considerable reversing effect of histopathologic and ultrastructural hypertensive nephropathy. Moreover, immunohistochemical results clearly pointed to relevant role of Nrf2 pathway in MSCs related renal therapeutic effects.

骨髓间充质干细胞(BM-MSCs)在再生医学领域引起了极大关注,人们开展了各种实验和临床试验来研究其对不同疾病的治疗效果。我们设计了一项组织学和免疫组织化学研究,以评估间充质干细胞疗法对高血压继发终末期肾病(ESRD)的治疗效果,高血压已成为一个日益严重且引人关注的公共卫生问题。我们使用了30只成年雄性白化大鼠,其中20只接受实验性高血压诱导,然后平均分为间充质干细胞治疗组(每只大鼠静脉注射1×10个荧光标记细胞),另一组不进行治疗。除了对血清尿素和肌酐进行生化测定外,还对肾标本进行了组织病理学、超微结构检查以及Nrf2免疫组织化学检查。我们的结果表明,骨髓来源的间充质干细胞对组织病理学和超微结构的高血压肾病具有显著的逆转作用。此外,免疫组织化学结果清楚地表明Nrf2通路在间充质干细胞相关的肾脏治疗效果中发挥了相关作用。
