To investigate the role of TGF-β1 on epithelial mesenchymal transition (EMT) and invasion in oral squamous cell carcinoma cell line Tca8113.
Cultured Tca8113 cells were treated with different concentrations of TGF-β1 for 24 h. The morphological changes were observed under phase-contrast microscopy. The mRNA and protein expression levels of EMT relative marker E-cadherin and Vimentin were detected by RT-PCR and Western blot. The effect of TGF-β1 on migration and invasion ability of Tca8113 cells were detected using transwell method.
The results demonstrated that TGF-β1 could induce morphological changes in Tca8113 cells from epithelial to mesenchymal. The mRNA and protein level of epithelial marker E-cadherin was downregulated following treatment with TGF-β1, whereas the mRNA and protein expression level of mesenchymal marker protein Vimentin was upregulated. Furthermore, TGF-β1 significantly enhances the migration and invasiveness of Tca8113 cells, which were effectively reversed by TGF-β1 inhibitor, LY2109761 CONCLUSIONS: TGF-β1 enhances Tca8113 cells migration and invasion by inducing epithelial mesenchymal transition.
探讨 TGF-β1 在口腔鳞状细胞癌细胞系 Tca8113 中的上皮间质转化(EMT)和侵袭中的作用。
用不同浓度的 TGF-β1 处理培养的 Tca8113 细胞 24 小时。相差显微镜下观察细胞形态变化。采用 RT-PCR 和 Western blot 检测 EMT 相关标志物 E-钙黏蛋白和波形蛋白的 mRNA 和蛋白表达水平。用 Transwell 法检测 TGF-β1 对 Tca8113 细胞迁移和侵袭能力的影响。
结果表明,TGF-β1 可诱导 Tca8113 细胞从上皮样向间质样形态变化。TGF-β1 处理后上皮标志物 E-钙黏蛋白的 mRNA 和蛋白水平下调,而间充质标志物蛋白波形蛋白的 mRNA 和蛋白表达水平上调。此外,TGF-β1 显著增强了 Tca8113 细胞的迁移和侵袭能力,TGF-β1 抑制剂 LY2109761 可有效逆转这一作用。
