Abstract

OBJECTIVE

In this study, we studied changes in histone acetylation during mouse oocytes meiosis. The aim was to investigate HDAC1 expression patterns in the mouse oocytes and effects of in-vitro maturation on epigenetic modifications during meiosis.

MATERIALS AND METHODS

Immature and mature oocytes were collected from female Kunming white mice of 4-6 weeks in age. Dynamic changes of histone H3K9, H4K12 acetylation were explored. HDAC1 spatial and temporal expression patterns during meiosis and their expression changes in in-vitro maturation were determined.

RESULTS

It was found that histone H3K9 and H4K12 acetylations were gradually disappeared during the meiotic maturation of mouse oocytes. HDAC1 proteins were localized mainly throughout the nucleoplasm in GV-intact oocyte, and colocalized with chromosomes at metaphase II (MII). The acetylated H3K9 and H4K12 were absent in oocytes matured in vivo, while the elevated acetylation of H3K9 and H4K12 was detected in oocytes matured in vitro. When cultured in vitro, the decrease of HDAC1 protein level and mRNA level were observed compared with oocytes matured in vivo.

CONCLUSIONS

the acetylation of H3K9, H4K12 decreased gradually to undetectable during oocyte meiosis. The histone deacetylation in oocytes was inadequate during in vitro maturation, and the in vitro maturation might lead to reduced HDAC1 expression in oocytes.

目的

本研究旨在探讨小鼠卵母细胞减数分裂过程中组蛋白乙酰化的变化。目的是研究 HDAC1 在小鼠卵母细胞中的表达模式以及体外成熟对减数分裂过程中表观遗传修饰的影响。

材料与方法

收集 4-6 周龄昆明小白鼠的未成熟和成熟卵母细胞。探索组蛋白 H3K9、H4K12 乙酰化的动态变化。确定 HDAC1 在减数分裂过程中的时空表达模式及其在体外成熟过程中的表达变化。

结果

研究发现,组蛋白 H3K9 和 H4K12 的乙酰化在小鼠卵母细胞减数成熟过程中逐渐消失。HDAC1 蛋白主要定位于 GV 期完整卵母细胞的核质中,并与中期 II 染色体(MII)共定位。体内成熟的卵母细胞中不存在乙酰化的 H3K9 和 H4K12,而体外成熟的卵母细胞中检测到 H3K9 和 H4K12 的乙酰化升高。与体内成熟的卵母细胞相比,体外培养时观察到 HDAC1 蛋白水平和 mRNA 水平下降。

结论

在卵母细胞减数分裂过程中,H3K9、H4K12 的乙酰化逐渐降低至无法检测。体外成熟过程中卵母细胞中的组蛋白去乙酰化不足,体外成熟可能导致卵母细胞中 HDAC1 表达减少。
