Abstract

Phytochemical investigation on the 70% EtOH extract of the leaves and branches of Gelsemium elegans resulted into the isolation of five new gelsedine-type oxindole alkaloids, gelseleganins A-E (1-5). The structures of the isolated compounds were established based on 1D and 2D (H-H COSY, HMQC, and HMBC) NMR spectroscopy, in addition to high resolution mass spectrometry. The isolated alkaloids were tested in vitro for cytotoxic potential against seven tumor cell lines. As a result, alkaloids 3 exhibited significant cytotoxic activities against all the tested tumor cell lines with IC values <10μM.

对钩吻叶和枝的70%乙醇提取物进行植物化学研究,从中分离出5种新的钩吻定型氧化吲哚生物碱,即钩吻素A - E(1 - 5)。除高分辨率质谱外,还基于一维和二维(氢-氢化学位移相关谱、异核多量子相干谱和异核多键相关谱)核磁共振光谱确定了分离出的化合物的结构。对分离出的生物碱进行体外测试,检测其对7种肿瘤细胞系的细胞毒性潜力。结果,生物碱3对所有测试的肿瘤细胞系均表现出显著的细胞毒性活性,其半数抑制浓度值<10μM。
