Abstract

BACKGROUND/AIM: Anthracimycin, a secondary metabolite of Streptomyces, has been shown to inhibit the invasion of certain cancer cell lines.

MATERIALS AND METHODS

In this study we evaluated the effect of anthracimycin on cell growth and signaling pathways in hepatocellular carcinoma (HCC).

RESULTS

Anthracimycin suppressed cell proliferation and motility and induced apoptosis in human HCC cell lines. Furthermore, anthracimycin had no effect on the enrichment of EpCAM-high liver cancer stem cells (CSCs), while fluorouracil dramatically enriched the CSCs with activation of the stemness-related genes EPCAM and SOX9 in HuH7 cells. Mechanistically, anthracimycin suppressed mammalian target of rapamycin (mTOR) signaling, and was most effective at inhibiting HCC cell proliferation with mTOR activation.

CONCLUSION

Anthracimycin is a novel mTOR inhibitor capable of suppressing the proliferation of CSCs and non-CSCs equally well in HCC, and it is suggested that anthracimycin could be effective in the eradication of HCC associated with mTOR-signaling activation.

背景/目的:蒽霉素是链霉菌的一种次级代谢产物,已被证明可抑制某些癌细胞系的侵袭。

材料与方法

在本研究中,我们评估了蒽霉素对肝癌(HCC)细胞生长和信号通路的影响。

结果

蒽霉素抑制人肝癌细胞系的细胞增殖和运动能力,并诱导其凋亡。此外,蒽霉素对EpCAM高表达的肝癌干细胞(CSCs)富集无影响,而氟尿嘧啶通过激活HuH7细胞中与干性相关的基因EPCAM和SOX9显著富集CSCs。机制上,蒽霉素抑制雷帕霉素靶蛋白(mTOR)信号传导,并且在抑制mTOR激活的肝癌细胞增殖方面最为有效。

结论

蒽霉素是一种新型的mTOR抑制剂,能够同等程度地抑制肝癌中CSCs和非CSCs的增殖,提示蒽霉素可能对根除与mTOR信号激活相关的肝癌有效。
