Abstract

OBJECTIVE

Histamine is an important chemical mediator in the development of allergic rhinitis and plays a key role in eliciting the nasal symptoms of the disorder. Histamine may also affect smell as a neurotransmitter. However, whether histamine receptors are present in the mammalian olfactory epithelium has not yet been examined. The aim of this study was to investigate the expression and distribution of histamine H, H, and H receptors in rat olfactory epithelium.

METHODS

Real-time quantitative PCR and immunohistochemical staining were performed to examine the mRNA level and protein expression and localization of histamine receptors (H, H, and H) in rat olfactory epithelium.

RESULTS

We demonstrated that mRNAs encoding histamine H, H, and H receptors were detected in rat olfactory epithelium. Immunohistochemistry also showed strong positive staining for these receptors. Co-localization of histamine H, H, and H receptors with olfactory mature protein revealed that these three histamine receptors were mainly localized in olfactory receptor neurons.

CONCLUSIONS

These findings indicate that histamine H, H, and H receptors are present in rat olfactory epithelium and may play a physiological role in olfactory transmission.

目的

组胺是变应性鼻炎发病过程中的一种重要化学介质,在引发该疾病的鼻部症状中起关键作用。组胺作为一种神经递质也可能影响嗅觉。然而,哺乳动物嗅觉上皮中是否存在组胺受体尚未得到研究。本研究的目的是调查组胺H1、H2和H3受体在大鼠嗅觉上皮中的表达及分布。

方法

采用实时定量PCR和免疫组织化学染色法检测大鼠嗅觉上皮中组胺受体(H1、H2和H3)的mRNA水平、蛋白表达及定位。

结果

我们证实在大鼠嗅觉上皮中检测到了编码组胺H1、H2和H3受体的mRNA。免疫组织化学也显示这些受体呈强阳性染色。组胺H1、H2和H3受体与嗅觉成熟蛋白的共定位显示,这三种组胺受体主要定位于嗅觉受体神经元。

结论

这些发现表明组胺H1、H2和H3受体存在于大鼠嗅觉上皮中,可能在嗅觉传导中发挥生理作用。
