A novel Gram-stain-negative, moderately halophilic, motile, facultatively anaerobic and rod-shaped strain, designated WDN1C137, was isolated from a marine saltern at Wendeng, PR China. Optimal growth occurred at 40 °C, pH 7.5 and with 7.0 % (w/v) NaCl. Q-10 was the sole respiratory quinone. The major cellular fatty acids (>10.0 %) in WDN1C137 were C18 : 1ω7c (46.2 %), cyclo C19 : 0ω8c (18.7 %) and C16 : 0 (12.3 %). The major polar lipids were phosphatidylglycerol, phosphoglycolipid, phosphatidylcholine, one unidentified glycolipid, one unidentified lipid, one unidentified aminolipid and two unidentified phospholipids. The genomic DNA G+C content was 70.9 mol%. Phylogenetic analysis based on 16S rRNA gene sequences revealed that WDN1C137 shared the highest similarity (94.5 %) to Roseivivax jejudonensis KCTC 42110, followed by Roseivivax halodurans JCM 10272 (94.2 %) and Roseivivax roseus DSM 23042 (94.1 %). WDN1C137 formed a separate branch from the closely related genera Roseivivax, Loktanella, Paracoccus and Cribrihabitans within the family Rhodobacteraceae, which indicated that it represented a novel genus in the phylogenetic tree. On the basis of the data from the current polyphasic study, the isolate is proposed to represent a novel species of a novel genus within the family Rhodobacteraceae, with the name Rhodosalinus sediminis gen. nov., sp. nov. The type strain of the type species is WDN1C137 (=KCTC 52478=MCCC 1H00170).
从中国山东省文登市的一个海盐场分离出一株革兰氏染色阴性、中度嗜盐、具运动性、兼性厌氧的杆状新菌株,命名为WDN1C137。该菌株在40 °C、pH 7.5和7.0 %(w/v)NaCl条件下生长最佳。泛醌Q-10是唯一的呼吸醌。WDN1C137中主要的细胞脂肪酸(>10.0 %)为C18 : 1ω7c(46.2 %)、环C19 : 0ω8c(18.7 %)和C16 : 0(12.3 %)。主要极性脂为磷脂酰甘油、磷酸糖脂、磷脂酰胆碱、一种未鉴定的糖脂、一种未鉴定的脂、一种未鉴定的氨基脂和两种未鉴定的磷脂。基因组DNA的G+C含量为70.9 mol%。基于16S rRNA基因序列的系统发育分析表明,WDN1C137与耶氏玫瑰色杆菌KCTC 42110的相似性最高(94.5 %),其次是嗜碱玫瑰色杆菌JCM 10272(94.2 %)和玫瑰色玫瑰色杆菌DSM 23042(94.1 %)。WDN1C137在红杆菌科内与近缘属玫瑰色杆菌属、洛氏菌属、副球菌属和栖海绵菌属形成一个独立分支,这表明它在系统发育树中代表一个新属。基于当前多相研究的数据,该分离株被提议代表红杆菌科内一个新属的一个新物种,命名为盐沼红杆菌属(Rhodosalinus),新属,新种。该新物种的模式菌株为WDN1C137(=KCTC 52478=MCCC 1H00170)。
