Abstract

OBJECTIVE

To observe the effect of electroacupuncture (EA) on the hormones derived from the hypothalamus-pituitary-ovary (HPO) axis, so as to explore the neuroendocrine mechanism induced by EA on rats with perimenopausal depression disorder.

METHODS

Sixty female sprague-dawley rats were randomly divided into blank control group, model group, sham-operation (sham) group, clomipramine group, and electroacupuncture (EA) group, with 12 rats in each group. Perimenopausal depression model was established by bilateral ovariectomy combined with chronic unpredictable stimulation.The EA group received continuous treatment at "Baihui" (GV 20), "Shenshu" (BL 23) and "Sanyinjiao" (SP 6) once a day for 28 days. Estrous cycle and sucrose preference test were monitored, and serum estradiol (E), luteinizing hormone (LH), gonadotropin releasing hormone (GnRH), and β-endorphin (β-EP) were detected by ELISA.

RESULTS

Compared to the blank control group, sugar water consumpution rates decreased in the model group and sham group (<0.05). Compared to the blank group and sham group, the serum LH and GnRH levels increased (<0.05), companied with lower serum E and β-EP levels in the model group (<0.05). Compared to the model group, sugar water consumpution rates increased in the clomipramine group and EA group (<0.05), both were companied with decreased serum LH and GnRH levels (<0.05), and higher serum E and β-EP levels (<0.05).

CONCLUSIONS

Electroacupuncture can relieve the symptoms of rat with perimenopausal depression by regulating the hormone secretion in HPO axis.

目的

观察电针(EA)对下丘脑-垂体-卵巢(HPO)轴衍生激素的影响,以探讨电针对围绝经期抑郁症大鼠的神经内分泌机制。

方法

将60只雌性Sprague-Dawley大鼠随机分为空白对照组、模型组、假手术(sham)组、氯米帕明组和电针(EA)组,每组12只。采用双侧卵巢切除术结合慢性不可预测刺激建立围绝经期抑郁模型。EA组于“百会”(GV 20)、“肾俞”(BL 23)和“三阴交”(SP 6)进行连续治疗,每天1次,共28天。监测动情周期和蔗糖偏好试验,并采用酶联免疫吸附测定法检测血清雌二醇(E)、黄体生成素(LH)、促性腺激素释放激素(GnRH)和β-内啡肽(β-EP)。

结果

与空白对照组相比,模型组和假手术组糖水消耗率降低(<0.05)。与空白组和假手术组相比,模型组血清LH和GnRH水平升高(<0.05),同时血清E和β-EP水平降低(<0.05)。与模型组相比,氯米帕明组和EA组糖水消耗率升高(<0.05),血清LH和GnRH水平均降低(<0.05),血清E和β-EP水平升高(<0.05)。

结论

电针可通过调节HPO轴激素分泌缓解围绝经期抑郁症大鼠的症状。
