This work aimed to evaluate the embryogenic competence of different tissues from different stages (friable callus, bud-regenerating callus, and whole buds) of , through the molecular detection of the somatic embryogenesis receptor-like kinase () and knotted-like homeobox () genes. RNA was extracted with Trizol, cDNA was obtained, and the studied transcripts were amplified. Using non-specific primers, and gene expression was detected in the three stages evaluated. This study might contribute to providing an explanation for the recalcitrance of this species to somatic embryogenesis.
这项工作旨在通过对体细胞胚胎发生受体样激酶(SERK)和类knotted同源框(KNOX)基因的分子检测,评估不同阶段(易碎愈伤组织、芽再生愈伤组织和完整芽)的不同组织的胚性能力。用Trizol提取RNA,获得cDNA,并扩增所研究的转录本。使用非特异性引物,在评估的三个阶段检测SERK和KNOX基因表达。本研究可能有助于解释该物种对体细胞胚胎发生的顽拗性。
