Abstract

OBJECTIVE

To study the effect of exogenous nitric oxide donor sodium nitroprusside (SNP) on antioxidant enzymes activities and lipid peroxidation of mice infected with .

METHODS

BALB/c mice were infected with separated by the digestion method. Forty-two days post-infection, the peripheral blood and hepatic tissue from the infected or normal mice were collected. Then 4 groups were set:liver homogenate from infected mice + SNP (Group A), liver homogenate from normal mice + SNP (Group B), peripheral blood from infected mice + SNP (Group C), and peripheral blood from normal mice + SNP (Group D). The final concentrations of SNP in each group were set as 0 (blank control), 2, 5, 10 μmol/L and 30 μmol/L, respectively. After reacting with SNP at 37 ℃ for 30 min, the superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) activities, and malondialdehyde (MDA) concentration were examined and compared.

RESULTS

The levels of SOD, CAT, GSH-Px and MDA concentration in the liver and the blood from the mice infected with were significantly higher than those of the normal ones (all < 0.05). When reacted with 10 μmol/L and 30 μmol/L SNP, the SOD, GSH-Px, and CAT activities in Group A and B decreased significantly (all < 0.05), while the liver MDA concentration reacted with 2-30 μmol/L SNP increased obviously (all < 0.05). As reacted with 30 μmol/L SNP, the activities of blood SOD, GSH-Px, and CAT in Group C and D decreased, while the MDA concentration in blood still increased (all < 0.01). When the SNP concentration was in the range of 2-30 μmol/L, there were a negative correlation between the SNP concentrations and SOD, GSH-Px, and CAT activities, as well as a positive correlation with the MDA concentration in the liver and blood from the mice infected with (all < 0.05).

CONCLUSIONS

infection could cause oxidative damage to mice, and increase SOD, GSH-Px, and CAT activities. Nitric oxide released from SNP can decrease antioxidase activities, and inhibit the antioxidant capacity of mice infected with .

目的

研究外源性一氧化氮供体硝普钠(SNP)对感染[具体感染物未明确]小鼠抗氧化酶活性及脂质过氧化的影响。

方法

采用消化法分离[具体感染物未明确]感染BALB/c小鼠。感染后42天,采集感染小鼠和正常小鼠的外周血及肝组织。然后设4组:感染小鼠肝匀浆+SNP(A组)、正常小鼠肝匀浆+SNP(B组)、感染小鼠外周血+SNP(C组)、正常小鼠外周血+SNP(D组)。每组SNP最终浓度分别设为0(空白对照)、2、5、10 μmol/L和30 μmol/L。在37℃与SNP反应30分钟后,检测并比较超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活性及丙二醛(MDA)浓度。

结果

感染[具体感染物未明确]小鼠肝脏和血液中的SOD、CAT、GSH-Px水平及MDA浓度均显著高于正常小鼠(均P<0.05)。与10 μmol/L和30 μmol/L SNP反应时,A组和B组的SOD、GSH-Px及CAT活性显著降低(均P<0.05),而与2 - 30 μmol/L SNP反应时肝脏MDA浓度明显升高(均P<0.05)。与30 μmol/L SNP反应时,C组和D组血液中SOD、GSH-Px及CAT活性降低,而血液中MDA浓度仍升高(均P<0.01)。当SNP浓度在2 - 30 μmol/L范围内时,感染[具体感染物未明确]小鼠肝脏和血液中SNP浓度与SOD、GSH-Px及CAT活性呈负相关,与MDA浓度呈正相关(均P<0.05)。

结论

[具体感染物未明确]感染可导致小鼠氧化损伤,并增加SOD、GSH-Px及CAT活性。SNP释放的一氧化氮可降低抗氧化酶活性,抑制感染[具体感染物未明确]小鼠的抗氧化能力。
