Abstract

BACKGROUND

Nasal mucosal epithelial cells express connexins, the prototypical gap junction proteins, and pannexins, a new family of channel proteins homologous to the invertebrate gap junction proteins. The physiological and pathophysiological roles of these transmembrane proteins in nasal mucosa are largely still unknown.

PURPOSE

Pannexins participate in ATP release into the extracellular space in various tissues, and ATP plays important roles in mucociliary clearance, especially by regulating ciliary beat activity. Therefore, we focused on the functional relationship between connexins, pannexin-1, ATP release, and mucociliary clearance in nasal epithelia.

RESULTS AND CONCLUSIONS

Connexins participate in the generation of intercellular calcium waves, in which calcium-mediated signaling responses spread to contiguous cells through the gap junction formed by connexins to transmit calcium signaling throughout the airway epithelium. Pannexins in the nasal mucosa may contribute to not only ciliary beat modulation via ATP release, but also regulation of mucus blanket components via HO efflux. The synchronized roles of pannexin and connexin may provide a new insight into effective mucociliary clearance systems in nasal mucosa.

目的

泛连接蛋白参与多种组织中ATP释放到细胞外空间的过程,且ATP在黏液纤毛清除中发挥重要作用,尤其是通过调节纤毛摆动活动。因此,我们重点研究了连接蛋白、泛连接蛋白-1、ATP释放与鼻上皮细胞黏液纤毛清除之间的功能关系。

结果与结论

连接蛋白参与细胞间钙波的产生,其中钙介导的信号反应通过连接蛋白形成的间隙连接扩散到相邻细胞,从而在整个气道上皮中传递钙信号。鼻黏膜中的泛连接蛋白可能不仅通过ATP释放有助于调节纤毛摆动,还通过H⁺外流调节黏液毯成分。泛连接蛋白和连接蛋白的协同作用可能为鼻黏膜中有效的黏液纤毛清除系统提供新的见解。
