A novel Gram-stain-negative, aerobic, non-spore-forming, motile and rod-shaped bacterial strain, designated HM451, was isolated from forest soil sampled at the Dinghushan Biosphere Reserve, Guangdong Province, PR China (112° 31' E 23° 10' N). It grew optimally at 28 °C, pH 5.0-6.0 and in the presence of 0-2.5 % (w/v) NaCl on R2A medium. Strain HM451 was closely related to Paraburkholderia mimosarum NBRC 106338 (98.6 % 16S rRNA gene sequence similarity), Paraburkholderia heleia NBRC 101817 (98.4 %) and Paraburkholderia silvatlantica SRMrh-20 (98.0 %). The 16S rRNA gene sequence analysis showed that strain HM451 and the three closely related strains formed a clade within the genus Paraburkholderia, but was clearly separated from the established species. The DNA-DNA relatedness value between strain HM451 and its phylogenetically closest relative, P. mimosarum NBRC 106338, was much lower than 70 %. Strain HM451 contained ubiquinone 8 as the major respiratory quinone. Major fatty acids were C16 : 0, C17 : 0cyclo and summed feature 8 (C18 : 1ω7c and/or C18 : 1ω6c). The DNA G+C content of strain HM451 was 65.4 mol%. The major polar lipids were phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, two unidentified aminophospholipids, one unidentified aminolipid and a polar lipid. The phenotypic, chemotaxonomic and phylogenetic data showed that strain HM451 represents a novel species of the genus Paraburkholderia, for which the name Paraburkholderia caseinilytica sp. nov. is proposed. The type strain is HM451 (=GDMCC 1.1190=LMG 30092).
从中国广东省鼎湖山生物圈保护区采集的森林土壤中分离出一株新型革兰氏阴性、需氧、无芽孢形成、具运动性的杆状细菌菌株,命名为HM451(东经112°31′,北纬23°10′)。它在R2A培养基上,于28°C、pH 5.0 - 6.0以及0 - 2.5%(w/v)NaCl存在的条件下生长最佳。菌株HM451与含羞草类伯克霍尔德菌NBRC 106338(16S rRNA基因序列相似性为98.6%)、赫氏类伯克霍尔德菌NBRC 101817(98.4%)和林地类伯克霍尔德菌SRMrh - 20(98.0%)密切相关。16S rRNA基因序列分析表明,菌株HM451与这三个密切相关的菌株在类伯克霍尔德菌属内形成一个分支,但明显与已确立的物种分开。菌株HM451与其系统发育关系最密切的相对菌株——含羞草类伯克霍尔德菌NBRC 106338之间的DNA - DNA相关性值远低于70%。菌株HM451含有泛醌8作为主要呼吸醌。主要脂肪酸为C16 : 0、C17 : 0环丙烷和总和特征8(C18 : 1ω7c和/或C18 : 1ω6c)。菌株HM451的DNA G + C含量为65.4 mol%。主要极性脂为磷脂酰乙醇胺、磷脂酰甘油、二磷脂酰甘油、两种未鉴定的氨基磷脂、一种未鉴定的氨基脂和一种极性脂。表型、化学分类和系统发育数据表明,菌株HM451代表类伯克霍尔德菌属的一个新物种,为此提出新名称酪解类伯克霍尔德菌(Paraburkholderia caseinilytica sp. nov.)。模式菌株为HM451(=GDMCC 1.1190 = LMG 30092)。
