Epidermal hyperproliferation with abnormal differentiation, inflammation, and angiogenesis are the key features of psoriasis. Glucose transporter-1 (GLUT-1) is a member of facilitative sugar transporters that are integral membrane glycoproteins moving sugar across cell membrane.
The objective of this study was to study the GLUT-1 expression in psoriasis.
Forty patients with psoriasis vulgaris and 20 healthy individuals were included in the study. Skin biopsies were taken from lesional and nonlesional skin of psoriasis patients as well as normal skin of control subjects. All were examined for GLUT-1 antibody expression by immunohistochemistry and GLUT-1 mRNA expression by real-time polymerase chain reaction (RT-PCR). In addition, specimens of psoriasis lesions were stained by hematoxylin and eosin and CD31 for morphometric analysis of histopathological parameters.
The intensity of GLUT-1 immunohistochemical expression and the relative levels of GLUT-1 mRNA expression in psoriasis lesions were upregulated in lesional skin of psoriasis patients in comparison with their nonlesional skin as well as normal control skin. GLUT-1 expression in psoriasis lesions showed significant positive correlations with Psoriasis Area and Severity Index (PASI) score, mean of epidermal thickness, inflammatory cell density, and microvessel density.
Glucose transporter-1 could play a role not only in the onset of psoriasis but also in the progression and severity of the disease. It may participate in the pathogenesis of psoriasis through the facilitation of epidermal hyperproliferation, inflammation, and angiogenesis.
表皮过度增生、异常分化、炎症和血管生成是银屑病的主要特征。葡萄糖转运蛋白-1(GLUT-1)是易化糖转运体的成员,是跨细胞膜移动糖的完整膜糖蛋白。
本研究旨在研究银屑病中 GLUT-1 的表达。
纳入 40 例寻常型银屑病患者和 20 例健康对照者。采集银屑病患者皮损和非皮损皮肤以及正常对照者的皮肤活检标本。采用免疫组织化学法检测 GLUT-1 抗体表达,实时聚合酶链反应(RT-PCR)检测 GLUT-1 mRNA 表达。此外,对银屑病皮损标本进行苏木精和伊红及 CD31 染色,对组织病理学参数进行形态计量学分析。
与非皮损皮肤和正常对照皮肤相比,银屑病患者皮损中 GLUT-1 免疫组化表达强度和 GLUT-1 mRNA 相对水平上调。银屑病皮损中 GLUT-1 表达与银屑病面积和严重程度指数(PASI)评分、表皮平均厚度、炎症细胞密度和微血管密度呈显著正相关。
葡萄糖转运蛋白-1 不仅在银屑病的发病中起作用,而且在疾病的进展和严重程度中也起作用。它可能通过促进表皮过度增生、炎症和血管生成参与银屑病的发病机制。
