Objective To investigate the effect of lycorine on the proliferation and apoptosis of gastric cancer SGC-7901 cells. Methods Effect of lycorine on the proliferation of SGC-7901 cells and 50% inhibitory concentration (IC) of lycorine were determined by CCK-8 assay. SGC-7901 cells were treated with IC lycorine, and 48 hours later, cell cycle and apoptosis were detected by flow cytometry, and the level of cyclin D1 mRNA and protein was detected by real-time fluorescence quantitative PCR and Western blot analysis. Results Lycorine obviously inhibited the proliferation of SGC-7901 gastric cancer cells in a concentration- or time-dependent manner. The IC of 24 hours and 48 hours to gastric cancer cells treated with lycorine was (20.85±2.36) μmol/L and (13.29±1.28) μmol/L, respectively. The cell cycle was arrested in G0/G1 phase, the apoptosis rate of cells increased, and the level of cyclin D1 mRNA and protein decreased significantly 48 hours after the treatment with 13 μmol/L lycorine. Conclusion Lycorine can inhibit the proliferation of SGC-7901 gastric cancer cells, induce cell cycle arrest in G0/G1 phase and promote cell apoptosis. The mechanism may be related to the down-regulation of cyclin D1 expression.
目的 探讨石蒜碱对胃癌SGC-7901细胞增殖和凋亡的影响。方法 采用CCK-8法检测石蒜碱对SGC-7901细胞增殖的影响及石蒜碱的半数抑制浓度(IC)。用IC石蒜碱处理SGC-7901细胞,48小时后,通过流式细胞术检测细胞周期和凋亡情况,通过实时荧光定量PCR和蛋白质免疫印迹分析检测细胞周期蛋白D1 mRNA和蛋白水平。结果 石蒜碱明显以浓度或时间依赖性方式抑制SGC-7901胃癌细胞的增殖。石蒜碱作用24小时和48小时对胃癌细胞的IC分别为(20.85±2.36)μmol/L和(13.29±1.28)μmol/L。细胞周期阻滞于G0/G1期,细胞凋亡率增加,用13 μmol/L石蒜碱处理48小时后,细胞周期蛋白D1 mRNA和蛋白水平显著降低。结论 石蒜碱可抑制SGC-7901胃癌细胞的增殖,诱导细胞周期阻滞于G0/G1期并促进细胞凋亡。其机制可能与细胞周期蛋白D1表达下调有关。
