To investigate the expression and clinical significance of miRNA-99a and miRNA-224 in the serum of patients with non-small cell lung cancer (NSCLC).
83 patients with NSCLC, who were diagnosed and treated in our hospital from January 2014 to September 2017, were included in the experiment group. 79 patients, who made health check up, were included in the control group. The quantitative Real Time-Polymerase Chain Reaction (qRT-PCR) technique was used to test the expressions of miRNA-99a and miRNA-224 in the serum of the patients in the two groups, and the relationship between the expression levels of miRNA-99a and miRNA-224 and the clinicopathological features of the patients with NSCLC was analyzed; the correlation between the expression of miRNA-99a and the expression of miRNA-224 in NSCLC was also analyzed.
The expression level of miRNA-99a in the patients with NSCLC was significantly lower than that in the patients in the normal control group; the differences were statistically significant (p<0.001). The expression level of miRNA-224 in the patients with NSCLC was markedly higher than that in the patients in the normal control group; the differences were statistically significant (p<0.001). The expression level of miRNA-99a in the patients with NSCLC was remarkably correlated with pathological stage, the presence or absence of lymph node metastasis and tissue differentiation (p<0.001). The expression level of miRNA-224 in the patients with NSCLC was significantly correlated with pathological stage, the presence or absence of lymph node metastasis and pathological grade (p<0.001). The results of partial correlation analysis showed that the expression levels of miRNA-99a and miRNA-224 were negatively correlated with each other in NSCLC (r=-0.985, p<0.001).
MiRNA-99a and miRNA-224 may be involved in the development and progression of NSCLC. MiRNA-99a is associated with NSCLC pathological stage, lymph node metastasis and tissue differentiation, while miRNA-224 is associated with NSCLC pathological stage, lymph node metastasis and pathological grade. MiRNA-99a and miRNA-224 can be used as clinical monitoring indicators for NSCLC.
研究微小 RNA-99a 和微小 RNA-224 在非小细胞肺癌(NSCLC)患者血清中的表达及临床意义。
选取 2014 年 1 月至 2017 年 9 月在我院诊断和治疗的 83 例 NSCLC 患者为实验组,选取同期健康体检者 79 例为对照组。采用实时荧光定量聚合酶链反应(qRT-PCR)技术检测两组患者血清中微小 RNA-99a 和微小 RNA-224 的表达,分析微小 RNA-99a 和微小 RNA-224 的表达水平与 NSCLC 患者临床病理特征的关系;分析 NSCLC 中微小 RNA-99a 与微小 RNA-224 的表达相关性。
与对照组相比,NSCLC 患者血清中微小 RNA-99a 的表达水平明显降低,差异具有统计学意义(p<0.001);与对照组相比,NSCLC 患者血清中微小 RNA-224 的表达水平明显升高,差异具有统计学意义(p<0.001)。NSCLC 患者血清中微小 RNA-99a 的表达水平与病理分期、有无淋巴结转移和组织分化程度显著相关(p<0.001);NSCLC 患者血清中微小 RNA-224 的表达水平与病理分期、有无淋巴结转移和病理分级显著相关(p<0.001)。偏相关分析结果显示,NSCLC 中微小 RNA-99a 和微小 RNA-224 的表达水平呈负相关(r=-0.985,p<0.001)。
微小 RNA-99a 和微小 RNA-224 可能参与 NSCLC 的发生和发展。微小 RNA-99a 与 NSCLC 病理分期、淋巴结转移和组织分化程度相关,微小 RNA-224 与 NSCLC 病理分期、淋巴结转移和病理分级相关。微小 RNA-99a 和微小 RNA-224 可作为 NSCLC 的临床监测指标。
