Abstract

OBJECTIVES

TObjectives: This study aims to investigate the effect of alendronate sodium on trabecular bone structure in an osteoporotic rat model.

MATERIALS AND METHODS

Between May 2006 and July 2006, 60 female Wistar Albino rats aged three months were randomly allocated to three groups: sham operated receiving no treatment (Shm); ovariectomized-alendronate receiving 1 mg/kg/day alendronate sodium (Ovx-A), and ovariectomized-vehicle receiving 1 mL/kg/day physiological saline (Ovx-PS). Both Ovx groups received treatment through gastric gavage for 56 days.

RESULTS

Densitometric measurements showed that bone mineral density decreased in the Ovx-PS and increased in Ovx-A groups (p<0.05). Biomechanical measurements showed a decrease in the breaking force in the Ovx-PS group and an increase in the Ovx-A group (p<0.05). Histomorphometric measurements showed that the Shm group had normal trabecular structure, while the Ovx-PS group had a less well- formed trabecular structure with a loss in the trabecular number and thickness and a corresponding increase in the trabecular spacing (p<0.05). In the Ovx-A group, there was an improvement in the trabecular structure with an increase in the trabecular number and thickness and a loss in the trabecular space (p<0.05).

CONCLUSION

Our study results suggest that alendronate sodium is a valuable treatment agent for osteoporosis in postmenopausal women.

目的

本研究旨在探讨阿仑膦酸钠对骨质疏松大鼠模型小梁骨结构的影响。

材料与方法

2006年5月至2006年7月,将60只3个月大的雌性Wistar白化大鼠随机分为三组:假手术未治疗组(Shm);卵巢切除-阿仑膦酸钠组,接受1mg/kg/天阿仑膦酸钠(Ovx-A);卵巢切除-赋形剂组,接受1mL/kg/天生理盐水(Ovx-PS)。两个卵巢切除组均通过灌胃给药56天。

结果

骨密度测量显示,Ovx-PS组骨矿物质密度降低,Ovx-A组骨矿物质密度增加(p<0.05)。生物力学测量显示,Ovx-PS组的断裂力降低,Ovx-A组的断裂力增加(p<0.05)。组织形态计量学测量显示,Shm组小梁结构正常,而Ovx-PS组小梁结构形成较差,小梁数量和厚度减少,小梁间距相应增加(p<0.05)。在Ovx-A组中,小梁结构得到改善,小梁数量和厚度增加,小梁间隙减小(p<0.05)。

结论

我们的研究结果表明,阿仑膦酸钠是绝经后妇女骨质疏松症的一种有价值的治疗药物。
