To investigate the role of prostate tumor overexpressed 1 (PTOV1) in the development and progression of human cervical cancer.
Real-time quantitative PCR, Western blot, and immunohistochemistry were used to explore PTOV1 expression in cervical cancer tissues and cell lines. Cell proliferation capability was examined by MTT assay. Statistical analyzes were applied to evaluate the correlation of PTOV1 expression with clinical parameters and prognosis.
The expression level of PTOV1 was markedly higher in cervical cancer tissues and cell lines than that in adjacent noncancerous tissues and the normal cervical epithelial cells. PTOV1 overexpression was correlated with higher tumor stage (P = 0.001), larger tumor size (P = 0.004), and lymph node involvement (P = 0.036). Moreover, patients with high PTOV1 expression showed shorter overall and recurrence-free survival time (P = 0.013 and P = 0.010, respectively). PTOV1 knockdown by short hairpin RNAi inhibited cancer cell growth in vitro.
PTOV1 may be an important factor associated with proliferation of cervical cancer.
探讨前列腺肿瘤过表达1(PTOV1)在人类宫颈癌发生发展中的作用。
采用实时定量聚合酶链反应、蛋白质免疫印迹法和免疫组织化学法检测宫颈癌组织及细胞系中PTOV1的表达。采用MTT法检测细胞增殖能力。应用统计学分析评估PTOV1表达与临床参数及预后的相关性。
宫颈癌组织及细胞系中PTOV1的表达水平明显高于相邻的非癌组织及正常宫颈上皮细胞。PTOV1过表达与更高的肿瘤分期(P = 0.001)、更大的肿瘤大小(P = 0.004)及淋巴结转移(P = 0.036)相关。此外,PTOV1高表达的患者总生存期和无复发生存期较短(分别为P = 0.013和P = 0.010)。短发夹RNA干扰敲低PTOV1可抑制体外癌细胞生长。
PTOV1可能是与宫颈癌增殖相关的重要因素。
