In order to determine six alkaloids (mass fraction) of nicotine, nornicotine, myosmine, anatabine, anabasine, and nicotyrine in tobacco and tobacco products quickly, accurately, and simultaneously, a novel method based on direct analysis of real-time model in situ ionization technique combined tandem mass spectrometry with a modified sample pretreatment was established, in which experimental parameters such as the type and amount of extraction solvent and injection rate were optimized, respectively. The samples of five commercial cigarettes and five kinds of tobacco leaves were analyzed by the established method, and the determined values were compared with those obtained using a gas chromatography with mass spectrometry method: (1) Under optimized conditions (30 mL ultrapure water as extraction solvent and with extraction rate of 0.6 mm/s), analysis could be completed within 10 min. (2) The linear range of the method was 0.002-2000 μg/g with , the recovery ranged from 86.8 to 105.6%, and the limit of detection and the limit of quantification were 0.004-0.835 μg/g and 0.013-2.787 μg/g, respectively. (3) The relative standard deviation between direct analysis of real-time method and the gas chromatography with mass spectrometry method was 0.34-8.83%. The established method is rapid, reliable, and suitable for the ultrafast determination of six alkaloids in tobacco and tobacco products.
为了快速、准确、同时测定烟草及烟草制品中的 6 种生物碱(尼古丁、降烟碱、新烟碱、假木贼碱、去甲烟碱和可替宁),建立了一种基于实时直接分析原位电离技术与串联质谱相结合的新型方法,对萃取溶剂的类型和用量、进样速率等实验参数进行了优化。采用所建立的方法对 5 种商业卷烟和 5 种烟叶样品进行分析,并与气相色谱-质谱法的测定值进行比较:(1)在优化条件下(30 mL 超纯水作为萃取溶剂,萃取速率为 0.6 mm/s),10 min 内可完成分析。(2)方法的线性范围为 0.002-2000 μg/g, , 回收率为 86.8%-105.6%,检出限和定量限分别为 0.004-0.835 μg/g 和 0.013-2.787 μg/g。(3)实时直接分析方法与气相色谱-质谱法之间的相对标准偏差为 0.34%-8.83%。所建立的方法快速、可靠,适用于烟草及烟草制品中 6 种生物碱的快速测定。
