Abstract

OBJECTIVE

The aim of this study was to evaluate the antiarthritic effects of different polar solvent extracts of Er Miao San (EMS) on model rats with adjuvant arthritis (AA) and screen the effective pats of EMS in the treatment of arthritis.

METHODS

Four different polar solvent extracts of EMS such as petroleum ether (PE), methylene chloride (CHCl), ethyl acetate (EtOAc), and -butanol (-butanol (.

RESULTS

Administration of EtOAc and CHCl parts remarkably inhibited the paw swelling, decreased the index of arthritis, decreased the body weight loss, and improved the changes of histopathology. Furthermore, the concentrations of proinflammatory cytokines (TNF-, IL-1, and IL-6) were significantly lower, while the anti-inflammatory cytokine (IL-10) was remarkably higher compared with that in the model group. And the result of UHPLC analysis indicated that the effective parts of EMS contain berberine and atractylodin.

CONCLUSIONS

EtOAc and CHCl are the effective parts of EMS that can improve arthritis. In particular, berberine and atractylodin may be responsible for the antiarthritic activity of EMS. This research provided pharmacological and chemical foundation for the application of EMS in treating rheumatoid arthritis (RA).

目的

本研究旨在评价二妙散(EMS)不同极性溶剂提取物对佐剂性关节炎(AA)模型大鼠的抗关节炎作用,并筛选EMS治疗关节炎的有效部位。

方法

制备EMS的四种不同极性溶剂提取物,如石油醚(PE)、二氯甲烷(CHCl)、乙酸乙酯(EtOAc)和正丁醇(-丁醇)。

结果

给予乙酸乙酯和二氯甲烷部位可显著抑制足爪肿胀,降低关节炎指数,减少体重减轻,并改善组织病理学变化。此外,与模型组相比,促炎细胞因子(TNF-、IL-1和IL-6)的浓度显著降低,而抗炎细胞因子(IL-10)显著升高。超高效液相色谱分析结果表明,EMS的有效部位含有小檗碱和苍术素。

结论

乙酸乙酯和二氯甲烷是EMS改善关节炎的有效部位。特别是,小檗碱和苍术素可能是EMS抗关节炎活性的原因。本研究为EMS在类风湿关节炎(RA)治疗中的应用提供了药理和化学基础。
