Multiple sclerosis (MS), one of the most common diseases of the central nervous system (CNS), is characterized by demyelination and chronic inflammation of the CNS. Failure of immune tolerance and induced autoimmune processes are involved in MS immunopathogenesis. Regulatory T (Treg) cells play an important role in maintaining peripheral tolerance and immune homeostasis.
The aim of this study was to evaluate the frequency of CD4CD25CD127 Treg cells in MS patients.
The study population was composed of 25 healthy controls (HCs), 35 patients with relapsing remitting multiple sclerosis (RRMS) and 25 patients with progressive multiple sclerosis (PMS). Frequency of CD4CD25CD127 Treg cells in RRMS and PMS patients was compared with HC by flow cytometry.
Treg cells frequency in PMS patients was significantly higher compared to RRMS patients ( < 0.001) and HCs ( < 0.001). It was lower in RRMS patients than HCs ( = 0.005). A Significant direct correlation between Treg cells frequency and expanded disability status scale (EDSS) in PMS patients ( = 0.001, = 0.6) was observed. Reverse correlation between Treg cells frequency and EDSS in RRMS patients was found ( = 0.01, = -0.4).
More de-tailed clarification of the role of Treg cells in MS patients could provide a basis for development of Treg cells-mediated therapeutic strategies.
多发性硬化症(MS)是中枢神经系统(CNS)最常见的疾病之一,其特征为CNS脱髓鞘和慢性炎症。免疫耐受失败及诱导的自身免疫过程参与了MS的免疫发病机制。调节性T(Treg)细胞在维持外周耐受和免疫稳态中起重要作用。
本研究旨在评估MS患者中CD4⁺CD25⁺CD127⁻ Treg细胞的频率。
研究人群包括25名健康对照者(HCs)、35名复发缓解型多发性硬化症(RRMS)患者和25名进展型多发性硬化症(PMS)患者。通过流式细胞术比较RRMS和PMS患者中CD4⁺CD25⁺CD127⁻ Treg细胞的频率与HCs。
PMS患者的Treg细胞频率显著高于RRMS患者(P<0.001)和HCs(P<0.001)。RRMS患者的Treg细胞频率低于HCs(P=0.005)。观察到PMS患者中Treg细胞频率与扩展残疾状态量表(EDSS)之间存在显著正相关(P=0.001,r=0.6)。在RRMS患者中发现Treg细胞频率与EDSS呈负相关(P=0.01,r=-0.4)。
对Treg细胞在MS患者中作用的更详细阐明可为开发Treg细胞介导的治疗策略提供依据。
