Abstract

OBJECTIVE

To study the behavioral changes of inositol 1, 4, 5-trisphosphate receptor type 2 knockout ( mice) and investigate the blood perfusion changes in the hippocampus using three-dimensional arterial spin labeling (3D-ASL).

METHODS

28 mice and 20 wild-type mice were assessed for depressive phenotype using behavioral tests (including sucrose consumption test, tail suspension test, forced swimming test and open field test). 15 mice and 14 wild-type mice were randomly selected for 3D-T2WI imaging of the whole brain and 3D-ASL imaging of the middle hippocampal layer, and cerebral blood flow (CBF) of the middle hippocampal layer was calculated. ITK-SNAP was used to delineate the bilateral hippocampal area and measure the average CBF value.

RESULTS

Compared with the wild-type mice, mice exhibited a distinct depressive phenotype with significantly decreased sucrose preference ( < 0.05) and increased immobile time in tail suspension test ( < 0.05) and forced swimming test ( < 0.01), without obvious changes in the performance in open field test ( > 0.05). Significantly decreased mean CBF values were found in the left and right hippocampus of mice as compared with the wild-type mice (left: 73.30 ±5.609 95.77±5.095; right: 73.53±5.700 100.5±4.696; bilateral means: 73.42±5.607 98.12±4.754; < 0.01).

CONCLUSIONS

Itpr2 deficiency can cause depressive phenotype and affect the cerebral blood flow in the hippocampus of mice.

目的

研究2型肌醇1,4,5-三磷酸受体敲除小鼠的行为变化,并采用三维动脉自旋标记(3D-ASL)技术研究海马区的血流灌注变化。

方法

采用行为学测试(包括蔗糖消耗试验、悬尾试验、强迫游泳试验和旷场试验)对28只敲除小鼠和20只野生型小鼠进行抑郁表型评估。随机选取15只敲除小鼠和14只野生型小鼠进行全脑三维T2加权成像(3D-T2WI)及海马中层三维动脉自旋标记成像(3D-ASL),计算海马中层的脑血流量(CBF)。使用ITK-SNAP软件勾勒双侧海马区域并测量平均CBF值。

结果

与野生型小鼠相比,敲除小鼠表现出明显的抑郁表型,蔗糖偏好显著降低(P<0.05),悬尾试验(P<0.05)和强迫游泳试验中的不动时间增加(P<0.01),旷场试验表现无明显变化(P>0.05)。与野生型小鼠相比,敲除小鼠左右海马的平均CBF值均显著降低(左侧:73.30±5.609对95.77±5.095;右侧:73.53±5.700对100.5±4.696;双侧平均值:73.42±5.607对98.12±4.754;P<0.01)。

结论

Itpr2基因缺陷可导致小鼠出现抑郁表型并影响海马区的脑血流量。
