Scorpion venom has toxic effects on mammals, insects and crustaceans. Toxicogenic peptides are major contributors to the scorpion venom, which make it toxic. The is one of the most common scorpion bites agent, and responsible for 95% of scorpion bite deaths cases in Iran.
In this project, we fractionated the scorpion venom and analyzed toxic fractions of the venom. The crude venom of was dialyzed against distilled water and then the soluble part of the venom was isolated from the non-soluble (mucoproteins) part of the venom and loaded onto the Sephadex G-50 gel filtration column, then after determining the toxicity of the obtained fractions (fractions toxicity were detected in mice by IV injection), the resulting toxic fraction was purified with three stages of ion-exchange chromatography (anion and cationic) and RP-HPLC. The purity of the fractions was verified by SDS-PAGE electrophoreses.
The LD of venom was 177.01 µg/mouse. The crude venom had 7 detectable bands with molecular weights of 10-100 KDa and one band less than 10 KDa. Finally, after the different stages of chromatography, two HL2153 and HL2155 peaks were obtained from the RP-HPLC, which were depicted single bands and high purity. The electrophoretic analysis showed molecular weight 4874 Da for HL2153 and 5107 Da for HL2155 toxins.
It is concluded that venom contains two HL2153 and HL2155 toxins with a relatively similar molecular weight and similar electrical charge 4874 and 5107 Da, respectively.
蝎毒对哺乳动物、昆虫和甲壳类动物具有毒性作用。产毒肽是蝎毒的主要成分,使其具有毒性。[蝎种名称]是最常见的蝎螫伤病原体之一,在伊朗95%的蝎螫伤死亡病例中都有它的身影。
在本项目中,我们对[蝎种名称]蝎毒进行了分级分离,并分析了其有毒成分。将[蝎种名称]的粗毒用蒸馏水透析,然后从毒液的不溶性(粘蛋白)部分分离出毒液的可溶性部分,并加载到Sephadex G - 50凝胶过滤柱上,在测定所得级分的毒性(通过静脉注射在小鼠中检测级分毒性)后,将所得有毒级分通过三个阶段的离子交换色谱(阴离子和阳离子)和反相高效液相色谱(RP - HPLC)进行纯化。通过SDS - PAGE电泳验证级分的纯度。
[蝎种名称]毒液的半数致死量(LD)为177.01 µg/小鼠。粗毒有7条可检测到的条带,分子量为10 - 100 KDa,还有一条小于10 KDa的条带。最后,经过不同阶段的色谱分离,从RP - HPLC中获得了两个HL2153和HL2155峰,它们呈现出单一条带且纯度高。电泳分析显示HL2153毒素的分子量为4874 Da,HL2155毒素的分子量为5107 Da。
得出的结论是,[蝎种名称]毒液含有两种HL2153和HL2155毒素,分子量相对相似,电荷分别为4874和5107 Da。
