Abstract

This study tested the possible protective and therapeutic effects of Viscum album extract and probiotics against carbon tetrachloride (CCl)-induced acute/chronic liver injury. Male Wistar rats were assigned to seven groups: Control, acute CCl, acute V. album + CCl, acute V. album + Probiotics + CCl, chronic CCl, chronic CCl + V. album, and chronic CCl + V. album + Probiotics. Acute and chronic liver injuries were induced by 2 mg/kg CCl (i.p.) and 1 mg/kg CCl (i.p.), respectively. The extract and probiotics were administered daily to related groups. Serum enzyme activities, lipid profile, total protein, albumin, bilirubin, heme oxgenase-1 and 8-hydroxydeoxyguanosine levels were measured. Liver tissue sections stained with Hematoxylin-Eosin. Acute or chronic CCl-exposure caused to significant changes in concentrations/activities of the measured parameters. The oral administration of extract and probiotics showed protective and therapeutic effects against CCl-induced liver-injury. The supplementation of intestinal flora by the use of probiotics may enhance the efficacy of orally given therapeutic extracts.

本研究测试了槲寄生提取物和益生菌对四氯化碳(CCl)诱导的急性/慢性肝损伤的可能保护和治疗作用。雄性 Wistar 大鼠被分为七组:对照组、急性 CCl 组、急性 CCl+槲寄生组、急性 CCl+益生菌+槲寄生组、慢性 CCl 组、慢性 CCl+槲寄生组和慢性 CCl+槲寄生+益生菌组。通过腹腔注射 2mg/kg CCl 和 1mg/kg CCl 分别诱导急性和慢性肝损伤。提取物和益生菌每日给予相关组。测量血清酶活性、脂质谱、总蛋白、白蛋白、胆红素、血红素加氧酶-1 和 8-羟基脱氧鸟苷水平。用苏木精-伊红对肝组织切片进行染色。急性或慢性 CCl 暴露导致所测量参数的浓度/活性发生显著变化。提取物和益生菌的口服给药对 CCl 诱导的肝损伤具有保护和治疗作用。益生菌对肠道菌群的补充可能增强口服给予的治疗性提取物的疗效。
