Abstract

OBJECTIVE

This study was designed to evaluate the effects of yixintongmai on proliferation, migration, and apoptosis of vascular smooth muscle cells (VSMCs) cultured with high glucose.

METHODS

VSMCs of the thoracic aorta from 5- to 8-week-old male Sprague-Dawley rats were cultured with normal (4.5 mM) or high (25 mM) glucose, respectively. The concentration of yixintongmai powder at 360 g/ml was chosen according to pre-experimental results.

RESULTS

Yixintongmai inhibited the proliferation of VSMCs (CCK-8 assay: 0.75 ± 0.04 versus 0.98 ± 0.09 OD, < 0.001; cell counting: 37533 ± 1861 versus 56009 ± 3779 cells/well, < 0.001) and the expression of proliferating cell nuclear antigen (0.74 ± 0.08 fold, < 0.001) as compared with high glucose (HG). Yixintongmai inhibited the migration of VSMCs (transwell assay: 146 ± 16 versus 265 ± 62 cells; < 0.001), scratch wound assay (0.17 ± 0.01 fold, < 0.001), and the expression of matrix metalloproteinases-9 (0.87 ± 0.03 fold, < 0.001) as compared with HG. Yixintongmai decreased mitochondrial membrane potentials (0.36 ± 0.12 fold, < 0.001) and promoted early (2.11 ± 0.20 fold, < 0.01) and late (2.11 ± 0.28 fold, < 0.01) apoptosis of VSMCs as compared with HG. Yixintongmai inhibited the expression of B-cell lymphoma 2 (0.83 ± 0.07 fold, < 0.01) and stimulated the activity of cleaved-capase-3/caspase-3 (2.00 ± 0.12 fold, < 0.05) as compared with HG. Yixintongmai inhibited reactive oxygen species generation (0.46 ± 0.03 fold, < 0.01) and the expression of NADPH oxidase-1 (0.84 ± 0.04 fold, < 0.001), nuclear factor-kappa B (NF-B) p65 (0.71 ± 0.07 fold, < 0.001), phosphorylated NF-B p65 (0.39 ± 0.02 fold,  < 0.0001), and inhibited nuclear translocation of NF-B p65 (0.87 ± 0.03 fold, < 0.001) in VSMCs as compared with HG.

CONCLUSIONS

Yixintongmai inhibits the proliferation and migration and promotes the apoptosis of VSMCs cultured with HG, which suggests the potential anti-atherosclerotic effects of this traditional Chinese medicine.

目的

本研究旨在评估益心通脉对高糖培养的血管平滑肌细胞(VSMCs)增殖、迁移及凋亡的影响。

方法

分别用正常(4.5 mM)或高糖(25 mM)培养5至8周龄雄性Sprague-Dawley大鼠胸主动脉的VSMCs。根据预实验结果选取益心通脉粉浓度为360 μg/ml。

结果

与高糖(HG)相比,益心通脉抑制VSMCs增殖(CCK-8法:0.75±0.04对0.98±0.09 OD,P<0.001;细胞计数:37533±1861对56009±3779个细胞/孔,P<0.001)及增殖细胞核抗原的表达(0.74±0.08倍,P<0.001)。与HG相比,益心通脉抑制VSMCs迁移(Transwell法:146±16对265±62个细胞,P<0.001)、划痕实验(0.17±0.01倍,P<0.001)及基质金属蛋白酶-9的表达(0.87±0.03倍,P<0.001)。与HG相比,益心通脉降低线粒体膜电位(0.36±0.12倍,P<0.001),促进VSMCs早期(2.11±0.20倍,P<0.01)和晚期(2.11±0.28倍,P<0.01)凋亡。与HG相比,益心通脉抑制B细胞淋巴瘤-2的表达(0.83±0.07倍,P<0.01),刺激裂解的半胱天冬酶-3/半胱天冬酶-3的活性(2.00±0.12倍,P<0.05)。与HG相比,益心通脉抑制活性氧生成(0.46±0.03倍,P<0.01)、NADPH氧化酶-1的表达(0.84±0.04倍,P<0.001)、核因子-κB(NF-κB)p65(0.71±0.07倍,P<0.001)、磷酸化NF-κB p65(0.39±0.02倍,P<0.0001),并抑制VSMCs中NF-κB p65的核转位(0.87±0.03倍,P<0.001)。

结论

益心通脉抑制HG培养的VSMCs的增殖和迁移,促进其凋亡,提示该中药具有潜在的抗动脉粥样硬化作用。
