This study is aimed at assessing the subsets of bone marrow macrophages in patients with myelodysplastic syndrome (MDS) and exploring the role of macrophages in the pathogenesis of MDS.
Thirty-eight newly diagnosed MDS patients were enrolled in the Department of Hematology of General Hospital of Tianjin Medical University from June 2015 to June 2016. Bone marrow monocytes and macrophage subsets (M1/M2) were detected in patients with MDS and normal controls by flow cytometry. M1 macrophages were cultured , and the expression of IL-1 and TNF- mRNA was measured using real-time polymerase chain reaction.
Compared with the normal control group, the proportion of bone marrow monocytes was higher (2.11 ± 0.93% vs. 3.66 ± 3.38%), and the mean fluorescence intensity of surface molecule CD14 was lower in the higher-risk (HR) MDS group (639.05 ± 359.78 vs. 458.26 ± 306.72, < 0.05). The ratio of M2 macrophages to monocytes was higher in patients with HR-MDS (1.82 ± 2.47% vs. 3.93 ± 3.81%, < 0.05). The ratio of M1 to M2 macrophages was lower in the HR-MDS group (3.50 ± 3.22 vs. 1.80 ± 0.88, < 0.05). The expression of IL-1 and TNF- mRNA in M1 macrophages was significantly lower in the MDS group ( < 0.05).
Patients with MDS had abnormal macrophage polarization, which may be involved in the alteration of bone marrow microenvironments.
本研究旨在评估骨髓巨噬细胞亚群在骨髓增生异常综合征(MDS)患者中的分布情况,并探讨巨噬细胞在 MDS 发病机制中的作用。
2015 年 6 月至 2016 年 6 月,天津医科大学总医院血液科收治 38 例初诊 MDS 患者,采用流式细胞术检测 MDS 患者和正常对照者骨髓单核细胞及巨噬细胞亚群(M1/M2)。培养 M1 巨噬细胞,采用实时聚合酶链反应检测白细胞介素-1(IL-1)和肿瘤坏死因子-α(TNF-α)mRNA 的表达。
与正常对照组比较,高危 MDS 组骨髓单核细胞比例较高(2.11±0.93%比 3.66±3.38%),表面分子 CD14 的平均荧光强度较低(639.05±359.78 比 458.26±306.72,<0.05)。HR-MDS 患者 M2 巨噬细胞与单核细胞的比值较高(1.82±2.47%比 3.93±3.81%,<0.05)。HR-MDS 组 M1 与 M2 巨噬细胞的比值较低(3.50±3.22 比 1.80±0.88,<0.05)。M1 巨噬细胞中 IL-1 和 TNF-αmRNA 的表达在 MDS 组明显降低(<0.05)。
MDS 患者存在巨噬细胞极化异常,这可能与骨髓微环境的改变有关。
