The role of bovine CD46 in the host cell entry of BVDV has been established for more than a decade. By generating novel MDBK CD46 knock-out clones, we confirm previously reported data on the CD46 motives important for BVDV binding and the importance of the G479R exchange within BVDV E to gain independence of bovine CD46 during entry. The comparison of different knock-out genotypes revealed a high variability of cellular susceptibility for a BVDV encoding the G479R exchange. These data highlight the effect of clonal selection of knock-outs on virus susceptibility, which should be considered when planning knock-out experiments.
十多年来,牛CD46在牛病毒性腹泻病毒(BVDV)进入宿主细胞过程中的作用已得到确认。通过构建新型的MDBK CD46基因敲除克隆,我们证实了先前报道的关于对BVDV结合重要的CD46基序的数据,以及BVDV E蛋白内G479R交换对于病毒进入过程中摆脱对牛CD46依赖的重要性。对不同基因敲除基因型的比较揭示了编码G479R交换的BVDV在细胞易感性方面存在高度变异性。这些数据突出了基因敲除克隆选择对病毒易感性的影响,在设计基因敲除实验时应予以考虑。
