This study aims to characterize the expression of ANGPTL4 in ovarian granulosa cells (GCs) and its association with polycystic ovary syndrome (PCOS).
This study included 104 PCOS patients and 112 women in control group undergoing fertilization-embryo transfer (IVF-ET) from the reproductive hospital affiliated with Shandong University from 2019 to 2021. By reverse transcription and real-time quantitative (RT-q) PCR, the mRNA expression of ANGPTL4 in GCs was assessed, and clinical information for these patients were then reviewed and analyzed.
The RT-qPCR results showed that ANGPTL4 expression in the control group was significantly lower than that in the PCOS group ( = 0.000) and had positive association with AMH ( = 0.211), HOMA-IR ( = 0.174), LDL/HDL ( = 0.176), ApoB/ApoAI ( = 0.155), and TC/HDL ( = 0.189). Additionally, the high expression of ANGPTL4 in the ovarian granulosa cells might be an independent predictor in PCOS (OR: 3.345; 95% CI: 1.951-5.734) with a close contact with incidence of PCOS (AUC: 0.704; 95% CI: 0.633-0.774, < 0.001).
Our study revealed higher ANGPTL4 expression in ovarian GCs with PCOS. Its association with glucose and lipid metabolism showed that ANGPTL4 might play an important role in PCOS metabolism and pathogenesis.
本研究旨在探讨 ANGPTL4 在卵巢颗粒细胞(GC)中的表达特征及其与多囊卵巢综合征(PCOS)的关系。
本研究纳入了 2019 年至 2021 年在山东大学附属医院生殖医院接受体外受精-胚胎移植(IVF-ET)的 104 例 PCOS 患者和 112 例对照组妇女。通过逆转录和实时定量(RT-q)PCR 评估 GC 中 ANGPTL4 的 mRNA 表达,并对这些患者的临床资料进行回顾性分析。
RT-qPCR 结果显示,对照组 ANGPTL4 的表达明显低于 PCOS 组( = 0.000),且与 AMH( = 0.211)、HOMA-IR( = 0.174)、LDL/HDL( = 0.176)、ApoB/ApoAI( = 0.155)和 TC/HDL( = 0.189)呈正相关。此外,卵巢颗粒细胞中 ANGPTL4 的高表达可能是 PCOS 的独立预测因子(OR:3.345;95%CI:1.951-5.734),与 PCOS 的发生率密切相关(AUC:0.704;95%CI:0.633-0.774, < 0.001)。
