Abstract

The mitochondrial unfolded protein response (UPR) is an evolutionarily conserved protective transcriptional response that maintains mitochondrial proteostasis by inducing the expression of mitochondrial chaperones and proteases in response to various stresses. The UPR-mediated transcriptional program requires the participation of various upstream signaling pathways and molecules. The factors regulating the UPR in Caenorhabditis elegans (C. elegans) and mammals are both similar and different. Cancer cells, as malignant cells with uncontrolled proliferation, are exposed to various challenges from endogenous and exogenous stresses. Therefore, in cancer cells, the UPR is hijacked and exploited for the repair of mitochondria and the promotion of tumor growth, invasion and metastasis. In this review, we systematically introduce the inducers of UPR, the biological processes in which UPR participates, the mechanisms regulating the UPR in C. elegans and mammals, cross-tissue signal transduction of the UPR and the roles of the UPR in promoting cancer initiation and progression. Disrupting proteostasis in cancer cells by targeting UPR constitutes a novel anticancer therapeutic strategy.

线粒体未折叠蛋白反应(UPR)是一种进化上保守的保护性转录反应,它通过响应各种应激诱导线粒体伴侣蛋白和蛋白酶的表达来维持线粒体蛋白质稳态。UPR介导的转录程序需要各种上游信号通路和分子的参与。秀丽隐杆线虫(线虫)和哺乳动物中调节UPR的因子既有相似之处,也有不同之处。癌细胞作为具有不受控制增殖的恶性细胞,面临来自内源性和外源性应激的各种挑战。因此,在癌细胞中,UPR被劫持并用于线粒体修复以及促进肿瘤生长、侵袭和转移。在本综述中,我们系统地介绍了UPR的诱导剂、UPR参与的生物学过程、线虫和哺乳动物中调节UPR的机制、UPR的跨组织信号转导以及UPR在促进癌症发生和进展中的作用。通过靶向UPR破坏癌细胞中的蛋白质稳态构成了一种新的抗癌治疗策略。
