To observe the effect of ZhuLian's type Ⅱ inhibition needling technique on sex hormones, insulin resistance and ovarian morphology in rats with polycystic ovary syndrome (PCOS), so as to explore its mechanism on regulating endocrine and metabolic dysfunction of PCOS.
Twenty-four female SD rats were randomly divided into control, model and ZhuLian acupuncture groups (=8 in each group). The PCOS model was established by continuous gavage of letrozole combined with high-fat diet. Rats of the ZhuLian acupuncture group were stimulated at "Guanyuan"(CV4) and bilateral "Guilai"(ST29), "Zusanli"(ST36), "Sanyinjiao"(SP6), and "Fenglong"(ST40) by ZhuLian's type Ⅱ inhibition needling technique for 30 min, continuously intervented for 28 days. The body weight was measured before and after modeling and after intervention. The ovarian volume was calculated. HE staining was used to observe the changes of ovarian histology. Plasma levels of luteinizing hormone (LH), follicle stimulating hormone (FSH) and fasting insulin (FINS) were measured by ELISA. LH/FSH ratio and insulin resistance index (HOMA-IR) were calculated.
After modeling, the rats' body weight and the ovarian volume were increased (<0.05), and the ovaries showed polycystic changes in the model group compared with the control group. And the FINS level, HOMA-IR, LH content and LH/FSH ratio were significantly increased (<0.05), while the FSH level was decreased (<0.05). After the intervention, the rats' body weight and ovarian volume were decreased (<0.05), and a small number of preantral follicles and more antral follicles were observed under microscope, and the levels of LH, LH/FSH, FINS and HOMA-IR were significantly decreased (<0.05), while FSH content was increased (<0.05) in the ZhuLian acupuncture group than in the model group.
ZhuLian's type Ⅱ inhibition needling technique can improve the endocrine and metabolic disorders in PCOS rats, which may be related to the regulation of endocrine system.
观察朱琏Ⅱ型抑制针法对多囊卵巢综合征(PCOS)大鼠性激素、胰岛素抵抗及卵巢形态的影响,以探讨其调节PCOS内分泌及代谢功能紊乱的机制。
将24只雌性SD大鼠随机分为对照组、模型组和朱琏针刺组(每组8只)。采用来曲唑连续灌胃联合高脂饮食建立PCOS模型。朱琏针刺组大鼠采用朱琏Ⅱ型抑制针法针刺“关元”(CV4)及双侧“归来”(ST29)、“足三里”(ST36)、“三阴交”(SP6)、“丰隆”(ST40),留针30分钟,连续干预28天。分别于造模前、造模后及干预后测量大鼠体重,计算卵巢体积。采用HE染色观察卵巢组织学变化。采用ELISA法检测血浆黄体生成素(LH)、卵泡刺激素(FSH)及空腹胰岛素(FINS)水平,计算LH/FSH比值及胰岛素抵抗指数(HOMA-IR)。
造模后,模型组大鼠体重及卵巢体积增加(<0.05),与对照组相比,卵巢呈现多囊样改变。且FINS水平、HOMA-IR、LH含量及LH/FSH比值显著升高(<0.05),而FSH水平降低(<0.05)。干预后,朱琏针刺组大鼠体重及卵巢体积减小(<0.05),显微镜下可见少量窦前卵泡及较多窦卵泡,与模型组相比,LH、LH/FSH、FINS及HOMA-IR水平显著降低(<0.05),而FSH含量升高(<0.05)。
朱琏Ⅱ型抑制针法可改善PCOS大鼠的内分泌及代谢紊乱,其机制可能与调节内分泌系统有关。
