Colitis associated colorectal cancer is a common intestinal malignant tumor, secondary to chronic enteritis. The persist existence of various inflammatory cells and cytokines in chronic inflammation promotes the development process of colitis-cancer transition. Interleukin 17 (IL-17) family, produced by Th17 cells, is found to be very important proinflammatory cytokines in various immune responses. In the proinflammatory response, IL-17 plays an important role in the activation and recruitment of neutrophils. Studies have shown the increased expression of IL-17 in serum and tumor tissues of patients with colorectal cancer. Th17 cells can induce the generation of immunosuppressive mediators including TGF-β, CXCR3, CCR6 and IL-6, which promotes the incidence of colorectal cancer. We summarized the role of IL-17 in the occurrence and progression of chronic colorectal inflammation to colorectal cancer, and elaborated the mechanisms by which IL-17 influences the formation of tumor microenvironment, promotes the proliferation, invasion and metastasis of tumor cells, and escapes immune surveillance.
结肠炎相关结直肠癌是一种常见的肠道恶性肿瘤,继发于慢性肠炎。慢性炎症中各种炎症细胞和细胞因子的持续存在促进了结肠炎-癌转化的发展过程。由Th17细胞产生的白细胞介素17(IL-17)家族在各种免疫反应中是非常重要的促炎细胞因子。在促炎反应中,IL-17在中性粒细胞的激活和募集中起重要作用。研究表明,结直肠癌患者血清和肿瘤组织中IL-17表达增加。Th17细胞可诱导包括转化生长因子-β(TGF-β)、CXC趋化因子受体3(CXCR3)、C-C趋化因子受体6(CCR6)和白细胞介素6(IL-6)等免疫抑制介质的产生,从而促进结直肠癌的发生。我们总结了IL-17在慢性结直肠炎症向结直肠癌发生发展中的作用,并阐述了IL-17影响肿瘤微环境形成、促进肿瘤细胞增殖、侵袭和转移以及逃避免疫监视的机制。
