Abstract

OBJECTIVE

The aim of the study is to investigate the therapeutic effects of astaxanthin (AST) and resveratrol (RVT) on multiorgan damage in an animal model of the supraceliac aortic ischemia-reperfusion (I/R).

METHODS

In this study, 28 rats ( = 7/group), 200 to 250 g in weight, were randomized to four groups (1: Sham, 2: Control + I/R, 3: AST + I/R, and 4: RVT + I/R). Following the abdominal incision, aortic dissection was performed in the sham group without injury. Other groups underwent I/R injury via supraceliac aortic clamping (20 minutes) and reperfusion. The rats were administered olive oil (3 mL/kg) orally for 2 weeks before and 1 week after the laparotomy. Additionally, oral AST (10 mg/kg) or RVT (50 mg/kg) was given to the study groups. All rats were sacrificed on the 3rd week of the experiment after blood samples were taken for analysis. Multiple rat tissues were removed.

RESULTS

We found that RVT increased total antioxidant status (TAS) and superoxide dismutase (SOD) levels, and decreased total oxidant status (TOS), oxidative stress index (OSI), myeloperoxidase (MPO), and malondialdehyde (MDA) levels, while AST increased the levels of TAS, decreased TNF-α, MDA, TOS, and OSI ( <0.05). Pathological investigations of the rat tissues revealed that both AST and RVT ameliorated tissue damage and apoptosis.

CONCLUSION

Our study suggests that AST and RVT might show therapeutic effects against oxidative tissue damage and apoptosis in an animal model of aortic I/R. Further studies are required.

目的

本研究旨在探讨虾青素(AST)和白藜芦醇(RVT)对肠系膜上动脉缺血再灌注(I/R)动物模型多器官损伤的治疗作用。

方法

本研究中,28 只大鼠(每组 7 只,体重 200-250g)随机分为四组(1:假手术组,2:对照组+I/R 组,3:AST+I/R 组和 4:RVT+I/R 组)。假手术组仅行剖腹切口,不进行主动脉夹层分离。其他组通过肠系膜上动脉夹闭(20 分钟)和再灌注进行 I/R 损伤。所有大鼠在剖腹手术后 2 周内和 1 周内口服橄榄油(3ml/kg)。此外,AST(10mg/kg)或 RVT(50mg/kg)口服给予研究组。实验第 3 周采血后处死所有大鼠,取多只大鼠组织。

结果

我们发现 RVT 增加了总抗氧化状态(TAS)和超氧化物歧化酶(SOD)水平,降低了总氧化状态(TOS)、氧化应激指数(OSI)、髓过氧化物酶(MPO)和丙二醛(MDA)水平,AST 则增加了 TAS 水平,降低了 TNF-α、MDA、TOS 和 OSI(<0.05)。大鼠组织病理学检查发现,AST 和 RVT 均能改善组织损伤和细胞凋亡。

结论

本研究表明,AST 和 RVT 可能对主动脉 I/R 动物模型中的氧化组织损伤和细胞凋亡具有治疗作用。需要进一步研究。
