Silica gel, octadecyl-silica(ODS), Sephadex LH-20, and semi-preparative high performance liquid chromatography(HPLC) was performed to isolate nine cephalotaxine-type alkaloids from Cephalotaxus sinensis: 8-oxodeoxyharringtonine(1), 8-oxonordeoxyharringtonine(2), cephafortunine A(3), 8-oxocephalotaxine(4), deoxyharringtonine(5), acetylcephalotaxine(6), cephalotaxine(7), epicephalotaxine(8), and cephalotaxinone(9). Compounds 1 and 2 were identified for the first time and their structures were determined by high-resolution-electrospray ionization-mass spectrometry(HR-ESI-MS), nuclear magnetic resonance(NMR), and electronic circular dichroism(ECD). Compounds 1-3 and 5 significantly inhibited the transcription of nuclear factor kappa B(NF-κB), with the half-maximal inhibitory concentration(IC_(50)) of(3.91±0.70),(2.99±0.45),(7.84±0.51), and(1.46±0.17) μmol·L~(-1), respectively.
采用硅胶、十八烷基硅胶(ODS)、葡聚糖凝胶LH - 20以及半制备高效液相色谱(HPLC)从中国粗榧中分离出9种三尖杉碱型生物碱:8 - 氧代脱氧三尖杉酯碱(1)、8 - 氧代去甲脱氧三尖杉酯碱(2)、三尖福林碱A(3)、8 - 氧代三尖杉碱(4)、脱氧三尖杉酯碱(5)、乙酰三尖杉碱(6)、三尖杉碱(7)、表三尖杉碱(8)和三尖杉酮碱(9)。化合物1和2为首次鉴定,其结构通过高分辨电喷雾电离质谱(HR - ESI - MS)、核磁共振(NMR)和电子圆二色光谱(ECD)确定。化合物1 - 3和5显著抑制核因子κB(NF - κB)的转录,其半数抑制浓度(IC₅₀)分别为(3.91±0.70)、(2.99±0.45)、(7.84±0.51)和(1.46±0.17)μmol·L⁻¹。
